Inhibition of 2,3-oxidosqualene cyclase by N-alkylpiperidines

Bioorganic & Medicinal Chemistry Letters, 1993
A series of N-alkylpiperidines (3)–(6), truncated A-ring analogs of the azadecalins (2) were synthesized, to further elucidate the structural requirements of OSC inhibition. From the structure-activity data obtained with the “clipped” A-ring analogs (3) and piperidines (4)–(6), important interactions between these inhibitors and oxidosqualene cyclase ...

Conjugated methyl sulfide and phenyl sulfide derivatives of oxidosqualene as inhibitors of oxidosqualene and squalene‐hopene cyclases

Lipids, 2003
AbstractVarious (1E,3E)‐ and (1Z,3E)‐conjugated methylthio derivatives of oxidosqualene (OS) and conjugated and non‐conjugated phenylthio derivatives of OS were obtained. These compounds, designed as inhibitors of pig liver and Saccharomyces cerevisiae 2,3‐oxidosqualene‐lanosterol cyclases (OSC) (EC 5.4.99.7) and of Alicyclobacillus acidocaldarius ...

2,3‐oxidosqualene cyclase: From azasqualenes to new site‐directed inhibitors

Lipids, 1995
Abstract2,3‐Oxidosqualene cyclases (OSC) are enzymes which convert 2,3‐oxidosqualene (OS) into polycyclic triterpenoids such as lanosterol, cycloartenol, and α‐and β‐amyrin. Our interest in the study of OSC is the development of new OSC inhibitors for potential use as hypocholesterolemic, antifungal, or phytotoxic drugs.
