Differential transcriptome analysis of glandular and filamentous trichomes in Artemisia annua [PDF]

open access: yes, 2013
Background: The medicinal plant Artemisia annua is covered with filamentous trichomes and glandular, artemisinin producing trichomes. A high artemisinin supply is needed at a reduced cost for treating malaria. Artemisinin production in bioreactors can be

A conserved amino acid residue critical for product and substrate specificity in plant triterpene synthases [PDF]

open access: yes, 2016
Triterpenes are structurally complex plant natural products with numerous medicinal applications. They are synthesized through an origami-like process that involves cyclization of the linear 30 carbon precursor 2,3-oxidosqualene into different triterpene

A Soluble 2,3-Oxidosqualene Sterol Cyclase

open access: yesJournal of Biological Chemistry, 1967
An enzyme catalyzing the anaerobic cyclization of 2,3-oxidosqualene into lanosterol has been extracted from hog liver microsomes by treatment with deoxycholate. Removal of deoxycholate, delipidation with ether, and ammonium sulfate precipitation yield a soluble 20-fold purified cyclase. No coenzyme requirements could be demonstrated for the cyclization

Oxidosqualene Cyclase (OSC) Inhibitors for the Treatment of Dyslipidemia

open access: yesCHIMIA, 2005
Novel inhibitors of oxidosqualene cyclase (OSC) for the treatment of dyslipidemia are reported. Starting point for the chemistry program was a set of compounds derived from a fungicide project which, in addition to high affinity for OSC from ...

The TriForC database : a comprehensive up-to-date resource of plant triterpene biosynthesis [PDF]

open access: yes, 2018
Triterpenes constitute a large and important class of plant natural products with diverse structures and functions. Their biological roles range from membrane structural components over plant hormones to specialized plant defence compounds.

Inactivation and activation of various membranal enzymes of the cholesterol biosynthetic pathway by digitonin.

open access: yesJournal of Lipid Research, 1989
The activity of rat liver microsomal squalene epoxidase is inhibited effectively by digitonin. Concentrations of 0.8 to 1.2 mg/ml of digitonin cause total inhibition of microsomal (0.75 mg protein/ml) squalene epoxidase either in microsomes that were ...

Integrated Metabolomic and Transcriptomic Analysis and Identification of Dammarenediol-II Synthase Involved in Saponin Biosynthesis in Gynostemma longipes

open access: yesFrontiers in Plant Science, 2022
Gynostemma longipes contains an abundance of dammarane-type ginsenosides and gypenosides that exhibit extensive pharmacological activities. Increasing attention has been paid to the elucidation of cytochrome P450 monooxygenases (CYPs) and UDP-dependent ...

Integrating pathway elucidation with yeast engineering to produce polpunonic acid the precursor of the anti-obesity agent celastrol

open access: yesMicrobial Cell Factories, 2020
Background Celastrol is a promising anti-obesity agent that acts as a sensitizer of the protein hormone leptin. Despite its potent activity, a sustainable source of celastrol and celastrol derivatives for further pharmacological studies is lacking ...

Effects of specific inhibition of sterol biosynthesis on the uptake and utilization of low density lipoprotein cholesterol by HepG2 cells.

open access: yesJournal of Lipid Research, 1991
Treatment of HepG2 cells in lipoprotein-deficient media with 4,4,10 beta-trimethyl-trans-decal-3 beta-ol (TMD) abolished the incorporation of [3H]acetate into cholesterol with concomitant accumulation of squalene 2,3(S)-oxide and squalene 2,3(S):22(S),23-

Identification of oxidosqualene cyclases associated with saponin biosynthesis from Astragalus membranaceus reveals a conserved motif important for catalytic function

open access: yesJournal of Advanced Research, 2023
Introduction: Triterpenoids and saponins have a broad range of pharmacological activities. Unlike most legumes which contain mainly oleanane-type scaffold, Astragalus membranaceus contains not only oleanane-type but also cycloartane-type saponins, for ...
