Intratumour heterogeneity complicates precision management of advanced endometrial cancer. Circulating tumor DNA (ctDNA) offers a minimally invasive strategy to capture tumor evolution and therapeutic resistance. Here, we compare tumor‐agnostic NGS with tumor‐informed ddPCR, outlining their relative sensitivity, concordance, and clinical implications ...

Interpreting the effects of DNA polymerase variants at the structural level
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Many patients with urothelial cancer do not benefit from treatment with pembrolizumab, while at risk of severe side effects. Changes in the levels of circulating tumor DNA early during treatment, measured by a simple and affordable assay that can be easily implemented in the clinic, can be used as a prognostic tool to identify these patients.

The novel styrylquinazolinone‐based molecule W1B effectively suppresses glioblastoma by inhibiting IGF1R and EGFR. In high‐glucose microenvironments driving tumor resistance, W1B acts synergistically with the EGFR inhibitor dacomitinib. This combination safely blocks compensatory survival signaling in zebrafish xenograft models. Showcasing promising in

Design optimization of permanent magnet actuators. [PDF]
This study describes the design optimization of permanent actuators, of both rotary and linear topologies. Parameter scanning, constrained single and multi-criterion optimization techniques are developed, with due emphasis on the efficient ...

Tumor B‐cell infiltration in platinum‐treated advanced muscle‐invasive urothelial carcinoma
Bladder tumors with higher pretreatment memory B‐cell infiltration were linked to longer survival after cisplatin chemotherapy, but not carboplatin. These tumors also showed more organized immune structures (tertiary lymphoid structures) and a shared pro‐inflammatory B‐cell‐rich community, suggesting that memory B cells may help identify patients most ...

On the adaptive selection of the parameter in stabilized finite element approximations [PDF]
A systematic approach is developed for the selection of the stabilization parameter for stabilized finite element approximation of the Stokes problem, whereby the parameter is chosen to minimize a computable upper bound for the error in the approximation.

The Optimal Saving with Mixed Parameters
This paper proposes two mixed models to study optimal saving in the presence of two types of risk: income risk and background risk. In the first model the income risk is a fuzzy number and the background risk is a random variable. In the second model the income risk is a random variable and the background risk is a fuzzy number.
