Phase II clinical trial of novel agent PBI-05204 in patients with metastatic pancreatic adenocarcinoma (mPDA).

Journal of Clinical Oncology, 2020
698 Background: Survival statistics for mPDA are dismal and with limited treatment options novel agents are needed to improve disease outcomes. PBI-05204 (Phoenix Biotechnology, Inc., San Antonio, TX) is a modified supercritical carbon dioxide extract of Nerium oleander leaves. Oleandrin, the extract’s major cytotoxic component, has demonstrated anti-

Abstract 5075: PBI-05204, a supercritical CO2 extract of Nerium oleander, suppresses the stemness of glioblastoma stem cells and inhibits the Shh-Gli1-NANOG axis

Cancer Research, 2022
Abstract Glioblastoma multiforme (GBM) is an aggressive form of primary brain neoplasm that has a poor prognosis and is resistant to conventional treatment. There is a great need to develop new strategies to improve the outcome of this disease.

A first-in-human phase I trial of PBI-05204 (oleandrin), an inhibitor of Akt, FGF-2, NF-Kb, and p70S6K in advanced solid tumor patients

Journal of Clinical Oncology, 2009
3537 Background: PBI-05204 contains oleandrin, a cardiac glycoside, which inhibits the α-3 subunit Na-K ATPase pump. Over-expression of the α-3 subunit in malignant cells correlates with the tumor proliferation. Oleandrin inhibits FGF- 2 export and activation of NF-Kβ and inhibits phosphorylation of Akt.

Final results of a first-in-human phase I trial of PBI-05204, an inhibitor of AKT, FGF-2, NF-Kb, and p70S6K in advanced cancer patients.

Journal of Clinical Oncology, 2011
3023 Background: PBI-05204 (Oleandrin), a cardiac glycoside, inhibits the α-3 subunit Na-K ATPase pump. Relative expression of the α-3 subunit in tumor cells correlates with proliferation. Oleandrin inhibits FGF-2 export, activation of NF-Kβ, phosphorylation of Akt, p70S6K, decreasing mTOR activity.

Abstract 5554: PBI-05204, a supercritical CO2 extract of Nerium oleander, inhibits the growth of human pancreatic cancer in a Panc-1 orthotopic model by down-regulation of PI3k/mTOR pathways.

Cancer Research, 2013
Abstract Recent studies have suggested that cardiac glycosides (CGs), such as oleandrin, have an ability to inhibit the proliferation of various cancer cells, including those from pancreatic, non-small cell lung cancer, prostate, colon and breast cancers.

PBI-05204, a supercritical CO₂ extract of Nerium oleander, inhibits growth of human pancreatic cancer via targeting the PI3K/mTOR pathway.

Investigational new drugs, 2016
Introduction Oleandrin, a cardiac glycoside, exerts strong anti-proliferative activity against various human malignancies in in vitro cells. Here, we report the antitumor efficacy of PBI-05204, a supercritical C0₂ extract of Nerium oleander containing oleandrin, in a human pancreatic cancer Panc-1 orthotopic model.
