Acidic pectin lyase is of great application value in fruit-processing industry. In this study, five pectin lyase genes (pelA, pelB, pelC, pelD and pelF) from A. niger pectin lyase gene family were cloned and over-expressed in A. niger strain SH-2, respectively.

Pectin-Degrading Enzymes: Polygalacturonase and Pectin Lyase
2005Pectic substances are polysaccharides rich in galacturonic acid. They are particularly abundant in the middle lamellae of plant tissues where they hold leaf cells together. During decomposition of leaf litter in streams, fungal activity softens leaf tissue and may release individual cells (maceration).

Publisher Summary This chapter discusses the synthesis of pectin lyase from fungi. The method of choice is based on the rate of formation of ∆4:5-unsaturated uronide product. Conjugation of the double bond with the C-6 carbonyl results in absorbance of ultraviolet light maximally at 235 mμ.

[Demonstration of pectin-lyase in Bacillus subtilis].
Comptes rendus de l'Academie des sciences. Serie III, Sciences de la vie, 1986After a screening performed on pectinolytic micro-organisms, a strain of B. subtilis was isolated. This strain has a pectic enzyme capable of beta-elimination on highly methylated pectin (degree of esterification = 85%), without the action of a pectinesterase. This activity corresponds to that of a pectin-lyase.

Carbohydrate Research, 2002Electrospray ionization (ESI) with quadrupole ion-trap mass spectrometry was used to assess the activity and specificity of the enzyme pectin lyase A (PLA) (EC 4.2.2.10) on model pectins with varying degrees and patterns of methyl esterification. PLA is a pectinase which cleaves alpha-(1-->4)-glycosidic linkages in pectin by a trans-elimination process.
