PERPRISE: A prospective non‐interventional study of PERampanel as only adjunctive treatment in patients with PRImary or SEcondarily generalized tonic–clonic seizures: First interim analysis

open access: yesEpilepsia Open
Objective To report the interim results of the PERPRISE study (Study 509; NCT04202159), which is evaluating perampanel as the only adjunctive anti‐seizure medication (ASM) in adults with focal to bilateral tonic–clonic seizures (FBTCS) or primary ...

Diagnostic Yield and Clinical Impact of Comprehensive WES/WGS Testing Beyond Common Genetic Causes in Hereditary Optic Atrophy

open access: yesClinical Genetics, EarlyView.
Opticus atrophy—Genetic testing with WES/WGS in 62 patients with optic atrophy provided a genetic diagnosis in 21 patients (33.9%). 42.9% of these involved non‐OPA1 genes, including WFS1, ACO2, NR2F1, UCHL1, CACNA1F, and COQ2, where the genetic diagnosis prompted additional clinical evaluation, surveillance, or therapeutic intervention.

HLA genotype testing for carbamazepine, oxcarbazepine and eslicarbazepine: A guideline developed by the UK Centre of Excellence in Regulatory Science and Innovation in Pharmacogenomics (CERSI‐PGx)

open access: yesBritish Journal of Clinical Pharmacology, Volume 92, Issue 7, Page 1957-1976, July 2026.
Carbamazepine is licensed in the United Kingdom for the treatment of epilepsy, bipolar disorder and trigeminal neuralgia. The related compounds oxcarbazepine and eslicarbazepine are licensed for the treatment of epilepsy. These drugs can cause immune‐mediated hypersensitivity reactions, which typically affect the skin, and can be of variable severity ...

Characterizing treatment initiation with central nervous system–active polypharmacy among adults with dementia

open access: yesAlzheimer's &Dementia, Volume 22, Issue 6, June 2026.
Abstract INTRODUCTION Central nervous system (CNS) –active polypharmacy, defined as concurrent use of antidepressants, antipsychotics, anti‐seizure medications, benzodiazepines, nonbenzodiazepine benzodiazepine receptor agonists, opioids, or skeletal muscle relaxants for ≥ 31 consecutive days, poses significant risks patients with dementia.

Severe Cutaneous Adverse Reactions Associated With Newer‐Generation Antiseizure Medications: A Real‐World Pharmacovigilance Study Based on FAERS and JADER

open access: yesCNS Neuroscience &Therapeutics, Volume 32, Issue 6, June 2026.
A cross‐database analysis of FAERS and JADER confirmed lamotrigine and zonisamide as the dominant SJS/TEN/DRESS risk drivers across both systems. After polytherapy adjustment, eslicarbazepine (DRESS, ROR 10.41) and levetiracetam emerged as reproducible novel signals warranting clinical attention. SJS/TEN onset concentrated within 4 weeks, whereas DRESS

Effectiveness and safety of cenobamate after conversion to monotherapy or dual therapy in real‐world clinical practice

open access: yesEpileptic Disorders, Volume 28, Issue 3, Page 775-785, June 2026.
Abstract Objective Patients with drug‐resistant epilepsy (DRE) typically take multiple anti‐seizure medications (ASMs) and are at risk of treatment‐related adverse events (AEs). This study assessed the impact of cenobamate monotherapy or dual therapy in patients with drug‐resistant epilepsy.
