ChromNet: A Multi‐Task Learning Framework for Cross‐Cell Type Prediction of 3D Chromatin Interactions Using Epigenetic Signals

open access: yesAdvanced Science, EarlyView.
ChromNet enables accurate cross‐cell‐type prediction of chromatin architecture from DNA and epigenetic profiles alone, capturing both conserved and cell‐type‐specific (including disease‐associated) structural features. Its noise‐perturbed multi‐task learning design enhances generalization, enabling precise reconstruction of 3D genome organization even ...

Type 2 Diabetes‐Associated Phenylacetylglutamine Induces Deleterious Inflammation Cycle in Myeloid Cells through β2 Adrenergic Receptors and Impedes Wound Healing

open access: yesAdvanced Science, EarlyView.
The microbiota‐derived metabolite phenylacetylglutamine (PAGln) is elevated in type 2 diabetes and tightly associated with poor healing in both diabetic and non‐diabetic human patients. PAGln promotes mouse inflammation and impairs healing through a transmissible β2‐adrenergic receptor–mediated trained‐immunity loop.

Single‐Cell RNA Sequencing of Thyroid Tissues Reveals Pathogenesis of Graves' Disease

open access: yesAdvanced Science, EarlyView.
Using single‐cell RNA sequencing and TCR profiling, this study constructed an immune cell atlas in thyroid tissue of Graves’ disease (GD), revealing dominant IFN‐γ‐secreting CD4+ T cells, expanded Tph and CD11c+ B cells, and enriched T/NK cells. Notably, GD thyroid follicular cells (TFCs) activated γδ T cells, which recruited cDC1 cells, suggesting a ...
