Inhibition of acyl‐CoA synthetase long‐chain isozymes decreases multiple myeloma cell proliferation and causes mitochondrial dysfunction

open access: yesMolecular Oncology, EarlyView.
Triacsin C inhibition of the acyl‐CoA synthetase long chain (ACSL) family decreases multiple myeloma cell survival, proliferation, mitochondrial respiration, and membrane potential. Made with Biorender.com. Multiple myeloma (MM) is an incurable cancer of plasma cells with a 5‐year survival rate of 59%.

Pharmaceutical Industry\u27s Effect on Socioeconomic Development in Sub-Saharan Africa in Relation to Family Planning Accessibility [PDF]

open access: yes, 2019
There is a large unmet need for family planning services in sub-Saharan Africa (Goodkind et al, 2018). Pharmaceutical companies contribute to the accessibility of medications in developing countries (Cottingham & Berer, 2011).

Hallmarks of Anaplastic Lymphoma Kinase Inhibitors with Its Quick Emergence of Drug Resistance

open access: yesPharmaceutical Fronts, 2022
Anaplastic lymphoma kinase (ALK) is one of the most popular targets for anticancer therapies. In the past decade, the use of anaplastic lymphoma tyrosine kinase inhibitors (ALK-TKIs), including crizotinib and ceritinib, has been a reliable and standard ...

Biomedical NER for the Enterprise with Distillated BERN2 and the Kazu Framework [PDF]

open access: yesarXiv, 2022
In order to assist the drug discovery/development process, pharmaceutical companies often apply biomedical NER and linking techniques over internal and public corpora. Decades of study of the field of BioNLP has produced a plethora of algorithms, systems and datasets.

TRPM4 contributes to cell death in prostate cancer tumor spheroids, and to extravasation and metastasis in a zebrafish xenograft model system

open access: yesMolecular Oncology, EarlyView.
Transient receptor potential melastatin‐4 (TRPM4) is overexpressed in prostate cancer (PCa). Knockout of TRPM4 resulted in reduced PCa tumor spheroid size and decreased PCa tumor spheroid outgrowth. In addition, lack of TRPM4 increased cell death in PCa tumor spheroids.

Enantioselective aza-Michael Cyclization Reaction Catalyzed by Quinine-Derived Monoquaternary Ammonium Salts: an Effective Route to Synthesize Letermovir

open access: yesPharmaceutical Fronts, 2021
A series of mono- or bis-quaternary ammonium salts derived from cinchonidine or quinine was synthesized and screened as potent phase-transfer catalysts for the reaction of aza-Michael cyclization, the key step in the synthesis of letermovir.

Respiratory complex I‐mediated NAD+ regeneration regulates cancer cell proliferation through the transcriptional and translational control of p21Cip1 expression by SIRT3 and SIRT7

open access: yesMolecular Oncology, EarlyView.
NAD+ regeneration by mitochondrial complex I NADH dehydrogenase is important for cancer cell proliferation. Specifically, NAD+ is necessary for the activities of NAD+‐dependent deacetylases SIRT3 and SIRT7, which suppress the expression of p21Cip1 cyclin‐dependent kinase inhibitor, an antiproliferative molecule, at the translational and transcriptional

Immunotoxicology in the Pharmaceutical Industry

open access: yesEnvironmental Health Perspectives, 1982
Development of an immunotoxicology program within the pharmaceutical industry is described. With few guidelines in the area and a multitude of factors to consider, a basic screen for evaluating immune competence in species routinely used in toxicologic studies has been proposed.

Peripheral blood leukocyte signatures as biomarkers in relapsed ovarian cancer patients receiving combined anti‐CD73/anti‐PD‐L1 immunotherapy in arm A of the NSGO‐OV‐UMB1/ENGOT‐OV30 trial

open access: yesMolecular Oncology, EarlyView.
Using mass cytometry, we analyzed serial blood samples from patients with relapsed epithelial ovarian cancer (EOC) treated with oleclumab–durvalumab combination immunotherapy in the NSGO‐OV‐UMB1/ENGOT‐OV30 trial. Our analysis identified potential predictive, monitoring, and response biomarkers detectable through liquid biopsy. These findings facilitate
