Phosphofructokinase-1 in Cancer: A Promising Target for Diagnosis and Therapy
Recent Patents on Anti-Cancer Drug DiscoveryAbstract: Tumor cells have distorted enzymatic houses, which change the metabolic state from oxidative phosphorylation to glycolysis with high lactate levels in a hypoxic environment. Redrafting the metabolic profile is an emerging hallmark of cancer. Glycolytic enzyme amplification occurs in about 70% of all malignancies.

Novel testis- and embryo-specific isoforms of the phosphofructokinase-1 muscle type gene
Biochemical and Biophysical Research Communications, 2004We have identified novel transcriptional isoforms of the human and mouse genes encoding muscle type phosphofructokinase-1 (PFK-M). These isoforms are expressed specifically in the testis and in the mid-gestation embryo, and have been termed TE-PFK-M (testis- and embryo-specific PFK-M).

Biopsies were obtained from the quadriceps femoris muscle of two male patients deficient in phosphofructokinase (PFK) 1. In the basal state the patients had markedly higher contents of UDP-glucose (approximately 5-fold), hexose monophosphates (approximately 7- to 13-fold), inosine monophosphate (IMP) (approximately 15-fold), and fructose 2,6 ...

Hepatic Phosphofructokinase-1 Activity and Fructose 2,6-Bisphosphate Levels in Patients with Abdominal Sepsis [PDF]
1. In sepsis various processes of carbohydrate metabolism, such as hepatic gluconeogenesis and glycolysis, are altered. Phosphofructokinase-1, a key glycolytic enzyme, is controlled in the long term via regulation of synthesis and degradation of the protein itself, while in the short term it is regulated by allosteric effectors, such as fructose 2,6 ...

Molecular cloning of phosphofructokinase 1 gene from a thermophilic bacterium, Thermus thermophilus
Biochemical and Biophysical Research Communications, 1991Phosphofructokinase 1 (PFK1) from Thermus thermophilus differs from other bacterial PFKs in that it is regulated by effector-induced reversible tetramer-dimer conversion rather than conformational change alone. We have cloned its gene and the deduced amino acid sequence was compared with that of other PFKs. While almost all amino acid residues involved

Phosphofructokinase-1 from Saccharomyces cerevisiae is composed of four alpha- and four beta-subunits, each of them carrying catalytic and regulatory bindings sites for MgATP. In this paper, various photoaffinity labels, such as 8-azidoadenosine 5'-triphosphate, 8-azido-1,N6-ethenoadenosine 5'-triphosphate, and 8-N3-3'(2')-O-biotinyl-8-azidoadenosine 5'
