Phosphorylase kinase isozymes and phosphorylase in denervated skeletal muscles

AbstractThe effects of motor denervation on levels of phosphorylase kinase isozymes and phosphorylase were investigated in rat epitrochlearis, hemidiaphragm, and soleus muscles. Amounts of the proteins were measured after quantitative immunoprecipitation and found to be decreased by as much as 70% 2 weeks after denervation.

Abstract A fully automated assay of phosphorylase kinase capable of processing 40 samples/hr is described. Problems due to enzyme adsorbed to the incubation coil and thereby producing a continuous rise of the base line were overcome by washing with a solution containing 0.05% Triton X-100, 0.03% SDS, and 5 m m EDTA.

Muscle phosphorylase b kinase (PHK) deficiency (glycogenosis type VIII) is a rare disorder caused by mutations in the PHKA1 gene encoding the alpha(M) subunit of PHK. Only 5 patients with molecular defects in the X-linked PHKA1 gene have been described until now, and they all presented with exercise intolerance.

A 35-year-old man had severe exercise intolerance and cramps. Venous blood lactate did not rise after ischemic exercise, and electromyographically silent contracture of hand muscles appeared. Histochemistry and electronmicroscopy of a muscle biopsy revealed subsarcolemmal and intermyofibrillar accumulation of glycogen.

Lymphocyte phosphorylase kinase activities in the sex-linked form of liver phosphorylase kinase deficiency

Lymphocyte phosphorylase kinase activities were measured in normal controls and in patients with the sex-linked form of liver phosphorylase kinase deficiency. The reaction due to phosphorylase kinase activity in normal lymphocytes (2.7 X 10(6) in the reaction tube) was found to be linear within 20-60 min at 30 degrees C.
