Harmonizing Rigidity and Flexibility: Embedding COF Quantum Dots Into Extracellular Matrix Gel as Carbon Monoxide Depot for Acoustically Triggered Time‐Programmable Anti‐Infective Therapy

open access: yesAdvanced Science, EarlyView.
A rigid‑flexible integrated CO depot is engineered by embedding rigid COFQDs into a flexible ECM gel network. The rigid framework enables high‑capacity CO storage and sonodynamic bacterial killing, while the flexible ECM gel controls sustained CO release, collectively eradicating multidrug‑resistant infection and promoting scarless healing.

Programmable Encapsulation Enables On‐Demand Proliferation of Therapeutic Bacteria for Potent Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
A programmable encapsulation technology is developed to reduce bacterial immunogenicity and proliferation after systemic administration. The cross‐linked polymer networks around individual bacteria enable immunogenic shielding and permit selective proliferation in tumors, allowing the bacteria to convert tumor‐accumulated ammonia into L‐arginine and ...

A Blood‐Derived Factor Rescues ALS: Platelet Factor 4 Activates OPTN‐Dependent Autophagy to Clear SOD1 Aggregates Independently of PINK1

open access: yesAdvanced Science, EarlyView.
Systemic platelet factor 4 (PF4) is significantly depleted in amyotrophic lateral sclerosis (ALS). Peripheral PF4 replenishment restores central proteostasis by driving OPTN‐dependent, PINK1‐independent selective autophagy in motor neurons. This intervention effectively clears toxic SOD1 aggregates, blunts glial activation, and preserves neuromuscular ...

Clinical study design and modeling approaches to study secretion of drugs in human milk

open access: yesFrontiers in Pediatrics
Breastfeeding has a multitude of benefits for both the lactating mother and her infant. However, lactating women often require medications for chronic and acute medical conditions, which may pose a significant risk due to drug transfer to infants via ...

Assessing Potential Drug–Drug Interactions Between Dabigatran Etexilate and a P-Glycoprotein Inhibitor in Renal Impairment Populations Using Physiologically Based Pharmacokinetic Modeling

open access: yes, 2019
Plasma concentrations of dabigatran, an active principle of prodrug dabigatran etexilate (DABE), are increased by renal impairment (RI) or coadministration of a P-glycoprotein inhibitor.

An Aggregation‐Induced Polymerization Poly(Disulfide)‐Drug Nanoplatform for Autoimmune Uveitis Therapy via Inhibiting the cGAS‐STING Pathway

open access: yesAdvanced Science, EarlyView.
A cationic poly(disulfide)‐drug nanoplatform (LA/DexP) was developed to treat experimental autoimmune uveitis (EAU). With potent blood‐retinal barrier penetrability, LA/DexP releases DSP in response to high ROS and scavenges cfDNA to inhibit the cGAS‐STING signaling pathway.

The Natural Product Corramycin Acts as a DNA Gyrase Poison and Overcomes Fluoroquinolone Resistance in Mycobacterium tuberculosis

open access: yesAdvanced Science, EarlyView.
Corramycin, a myxobacterial natural product antibiotic, exhibits potent bactericidal activity against multidrug‐resistant Mycobacterium tuberculosis. The compound hijacks bacterial transporters such as BacA and OppABCD to enter the cell and inhibits DNA synthesis through a previously unrecognized mode of DNA gyrase poisoning, locking the enzyme in an ...
