Applications of physiologically based pharmacokinetic modeling for the optimization of anti-infective therapies [PDF]

open access: yes, 2015
Introduction: The pharmacokinetic properties of anti-infective drugs are a determinant part of treatment success. Pathogen replication is inhibited if adequate drug levels are achieved in target sites, whereas excessive drug concentrations linked to ...

Physiologically Based Pharmacokinetic Modeling of Therapeutic Proteins [PDF]

open access: yesJournal of Pharmaceutical Sciences, 2017
Biologics or therapeutic proteins are becoming increasingly important as treatments for disease. The most common class of biologics are monoclonal antibodies (mAbs). Recently, there has been an increase in the use of physiologically based pharmacokinetic (PBPK) modeling in the pharmaceutical industry in drug development.

A Single‐Metal‐Doped Nanoplatform for Ferroptosis‐Driven cGAS‐STING Pathway Activation in Hepatocellular Carcinoma Immunotherapy

open access: yesAdvanced Functional Materials, EarlyView.
The cGAS‐STING pathway boosts HCC antitumor immunity but lacks specific activation. Nanoplatform ZMRPF induces HCC ferroptosis via lipid ROS, releasing mtDNA. It synergizes with ZMRPF‐released Mn2⁺ to activate cGAS‐STING, amplifies antigen‐presenting cell activity, reverses HCC immunosuppression, and enables robust systemic antitumor immunity ...

Addressing health equity for breastfeeding women: primaquine for Plasmodium vivax radical cure

open access: yesMalaria Journal
Plasmodium vivax malaria remains a global health challenge, with approximately 6.9 million estimated cases in 2022. The parasite has a dormant liver stage, the hypnozoite, which reactivates to cause repeated relapses over weeks, months, or years.

Mechanical and Electrical Phenotype of hiPSC‐Cardiomyocytes on Fibronectin‐Based Hydrogels

open access: yesAdvanced Healthcare Materials, EarlyView.
We introduce fibronectin‐based PEG hydrogels with controlled rigidity to enable the culture of iPSC‐derived cardiomyocytes. These substrates offer an alternative to the current culture of these cells on fibronectin‐coated glass, providing enhanced structural and functional behavior. The system provides a more physiologically relevant platform to assess

Interaction of Rifampin and Darunavir-Ritonavir or Darunavir-Cobicistat In Vitro [PDF]

open access: yes, 2017
Treatment of HIV-infected patients coinfected with Mycobacterium tuberculosis is challenging due to drug-drug interactions (DDIs) between antiretrovirals (ARVs) and antituberculosis (anti-TB) drugs.

Photoactivated Proximity Protein Labeling Reveals Enhanced Tumor Retention of a D‐Peptide‐Ruthenium Prodrug Conjugate

open access: yesAdvanced Healthcare Materials, EarlyView.
Six stereoisomeric ruthenium‐peptide conjugates were synthesized and characterized, which showed high potential in photoactivated chemotherapy (PACT) and proximity protein labeling. Abstract Amino acid chirality is known to influence the biological properties of peptide‐containing prodrugs.

Metallofullerenol Sc3N@C80(OH)18: A New Generation Radioprotector Protecting Human Erythrocytes Against Multiple Biochemical Damage Modes Upon Gamma Irradiation, Identifying It as a Scavenger of Short‐ and Long‐Lived Radicals

open access: yesAdvanced Healthcare Materials, EarlyView.
Metallofullerenol Sc3N@C80(OH)18 demonstrates strong radioprotective properties as a scavenger of both short‐ and long‐lived radicals. The study reveals protection of human erythrocytes from γ‐radiation–induced biochemical damage via post‐irradiation removal of primary and secondary reactive oxidants, supported by pulse radiolysis kinetics.
