Kinetic studies on the phosphorolysis of polynucleotides by polynucleotide phosphorylase

open access: yesJournal of Biological Chemistry, 1975
The kinetics of the phosphorolysis of polynucleotide (as differentiated from oligonucleotide) by polynucleotide phosphorylase of Micrococcus luteus has been investigated. Double reciprocal plots of initial velocity against either inorganic phosphate or polynucleotide concentration are linear, and furthermore, the affinity of the enzyme for either ...

Driven translocation of a polynucleotide chain through a nanopore--A continuous time Monte-Carlo study [PDF]

open access: yesPhysical Review E74, 011911 (2006), 2006
Using continuous time Monte-Carlo method we simulated the translocation of a polynucleotide chain driven through a nanopore by an electric field. We have used two models of driven diffusion due to the electric field. The chain may have strong interaction with the pore, and depends on which end of the chain first enters the pore.

Translocation of structured polynucleotides through nanopores [PDF]

open access: yesPhysical Biology 1 (2004) 19-26, 2003
We investigate theoretically the translocation of structured RNA/DNA molecules through narrow pores which allow single but not double strands to pass. The unzipping of basepaired regions within the molecules presents significant kinetic barriers for the translocation process.

Molecular dynamics study on the mechanism of polynucleotide encapsulation by chitosan

open access: yesScientific Reports, 2017
The safe and effective delivery of therapeutic genes into target cell interiors is of great importance in gene therapy. Chitosan has been extensively studied as a gene delivery carrier due to its good biocompatibility and biodegradability.

Synthesis of Polynucleotide Analogs Containing a Polyvinyl Alcohol Backbone

open access: yesMolecules, 2008
Water soluble homo-base polynucleotide analogues were synthesized in whichpolyvinyl alcohol and partially phosphonated polyvinyl alcohol constituted the backbones, onto which were grafted uracil or adenine via 1,3-dioxane spacers formed by acetalformation

Fluorescent labeling of plasmid DNA and mRNA : gains and losses of current labeling strategies [PDF]

open access: yes, 2016
Live-cell imaging has provided the life sciences with insights into the cell biology and dynamics. Fluorescent labeling of target molecules proves to be indispensable in this regard. In this Review, we focus on the current fluorescent labeling strategies

Stochastic sensing of polynucleotides using patterned nanopores [PDF]

open access: yesPhys. Rev. X 2, 021002 (2012), 2012
The effect of the microscopic structure of a pore on polymer translocation is studied using Langevin dynamics simulation, and the consequence of introducing patterned stickiness inside the pore is investigated. It is found that the translocation process is extremely sensitive to the detailed structure of such patterns with faster than exponential ...

Synthesis, Cyclopolymerization and Cyclo-Copolymerization of 9-(2-Diallylaminoethyl)adenine and Its Hydrochloride Salt

open access: yesMolecules, 2012
We report herein the synthesis and characterization of 9-(2-diallylaminoethyl) adenine. We evaluated two different synthetic routes starting with adenine where the optimal route was achieved through coupling of 9-(2-chloroethyl)adenine with diallylamine.

Impact of PNKP mutations associated with microcephaly, seizures and developmental delay on enzyme activity and DNA strand break repair [PDF]

open access: yes, 2012
Microcephaly with early-onset, intractable seizures and developmental delay (MCSZ) is a hereditary disease caused by mutations in polynucleotide kinase/phosphatase (PNKP), a DNA strand break repair protein with DNA 5'-kinase and DNA 3'-phosphatase ...
