Resistance to the Potato Leafroll Virus (PLRV) in Diploid Potatoes

Plant Breeding, 1989
AbstractA high level of PLRV resistance has been found in four diploid genotypes originating from resistant ancestors widely utilized in European potato breeding. Plants of these genotypes were difficult to infect not only with aphids, but also with graft inoculation.

Reaction to the potato leafroll virus (PLRV) in diploid potatoes

Potato Research, 1988
Diploid parents with some resistance to PLRV, were intercrossed to give 3 families with 191 clones which were evaluated for reaction to PLRV and yielding ability. After inoculation with PLRV the clones could be separated into those: 1) resistant, 2) susceptible, 3) intolerant, reacting with low virus concentration, 4) tolerant and 5) intermediate in ...

Incidence of Potato Viruses S and X and Potato Leafroll Virus in Potatoes in Queensland.

Australasian Plant Pathology, 1988
In 1986-87, enzyme-linked immunosorbent assay (ELISA) was used to test for potato virus S (PVS), potato virus X PVX) and potato leafroll virus (PLRV) in certified potato "seed" obtained from either Victoria or New South Wales. one of the viruses was found in the Victorian "seed", but one out of three samples from New South Wales contained 10% PVS.

A characterization of epitopes on potato leafroll virus coat protein

Archives of Virology, 1990
A panel of ten stable hybridoma cell lines secreting monoclonal antibodies (MAbs) specific for potato leafroll virus (PLRV) antigen, was produced in two fusion experiments with murine splenic and myeloma cells. Using different ELISA procedures and Western blotting it was shown that one MAb detected a continuous epitope and nine MAbs reacted with ...

Cloning of the gene for the capsid protein of potato leafroll virus

Archives of Virology, 1989
DNA complementary to the RNA of purified potato leafroll virus (PLRV) was synthesized and cloned into the lambda insertion vector NM1149. Overlapping PLRV-specific cDNA clones were isolated that represent some 80% of the viral genome. Sequences coding for the capsid protein were identified by subcloning size-selected cDNAs into the lambda expression ...

Characterization of potato leafroll virus

2010
Potato leafroll virus (PLRV) was purified from infected potato (Splanum tuberosum L.) by using a mixture of chloroform and butanol for clarification and polyethylene glycol for concentration. The virus sedimented as a single band in a sucrose density gradient. Yield of purified virus varied from 0.4-0.6 mg/kg fresh weight of potato foliage. Virus yield

Multiple components of the resistance of potatoes to potato leafroll virus

Annals of Applied Biology, 1987
SUMMARYIn glasshouse experiments the ranking of potato genotypes for resistance to infection with potato leafroll virus (PLRV) using three concentrations of aphid‐borne inoculum was the same as their field resistance ratings. In field‐grown plants this resistance to infection increased in all genotypes as the plants aged but its rate of increase ...

Restricted multiplication of potato leafroll virus in resistant potato genotypes

Annals of Applied Biology, 1985
SUMMARYPlants of a range of potato genotypes differing in rating for field resistance to potato leafroll virus (PLRV) were inoculated with the virus by grafting or by aphids (Myzus persicae). Plants of all genotypes tested became infected by each inoculation method and PLRV was detected by ELISA in the upper leaves of all genotypes within 26 days after

Potato leafroll virus contains a double-stranded DNA

Virology, 1976
Abstract The nucleic acid of potato leafroll virus (PLRV) has been isolated from highly purified virus preparations by two methods: (1) From a treatment with 0.5 N perchloric acid at 70° and quantitative estimation of the products, the DNA:protein ratio has been found to be nearly 40:60 (w/w).

Analysis of epitopes on potato leafroll virus capsid protein

Virology, 1992
Pepscan hexapeptides prepared to the capsid protein amino acid sequence of potato leafroll luteovirus (PLRV) were tested against both polyclonal and monoclonal antibodies. Twelve continuous epitopes were identified: 11 were detected by two different PLRV polyclonal antisera, but only 4 were detected by both antisera.
