Potato leafroll virus (PLRV) and potato spindle tuber viroid (PSTVd) are important pathogens affecting the potato (Solanum tuberosum L.). PLRV is transmitted only by aphids, Myzus persicae being its principal vector. In contrast, PSTVd is transmitted by foliar contact and true seeds, but is not aphid‐transmissible.

Purification and characterization of potato leafroll virus

Potato leafroll virus (PLRV) was purified from infected potato (Solanum tuberosum L.) with yields of 0.4-0.6 mg/kg of foliage. The virus sedimented as a single component of 127 S. An antiserum prepared against purified virus had a maximum titer of 1:1024 in agar gel double diffusion tests.

Ultrastructure of potato leaf phloem infected with potato leafroll virus

Electron microscopy of Russet Burbank potato (Solanum tuberosum L.) leaf midveins infected with the potato leafroll virus (PLRV) revealed the pathological effects induced by the virus in the phloem. Virus particles were observed in mature sieve elements, companion cells, and plasmodesmata connecting the two kinds of cells.

Host range of potato leafroll virus

The susceptibility to potato leafroll virus of over 150 species distributed among 39 families was determined. Inoculations were made by means of the peach aphid,Myzus persicae. The species considered susceptible are those from which the virus could be recovered 3 to 6 weeks following inoculation.
