In search of preclinical robustness
Nature Reviews Drug Discovery, 2012Systematic engagement of statisticians in preclinical research could help address the weaknesses that are undermining the likelihood of subsequent success in drug discovery and development.

Preclinical data for Droloxifene
Cancer Letters, 1994The new antiestrogen Droloxifene has a 10-60-fold higher binding affinity to the estrogen receptor (ER) compared to the related compound Tamoxifen. A similar relationship was found in growth inhibition studies which showed that Droloxifene inhibited the different ER positive human breast cancer cells more effectively than Tamoxifen, predominantly in ...

Surgical Infections, 2012
A variety of sepsis models have been used to unravel pathophysiologic processes and to examine the effects of novel therapeutic interventions. The lack of therapeutic efficacy of numerous compounds in clinical sepsis trials, despite glorious results in animal models of sepsis, has raised doubt and debate about the usefulness of such models.Review of ...

Preclinical pharmacology of sugammadex
Journal of Critical Care, 2009Since the introduction of nondepolarizing neuromuscular blocking agents, acetylcholinesterase inhibitors have been used to increase the speed of recovery from neuromuscular blockade. The major disadvantages of acetylcholinesterase inhibitors are their lack of activity against profound neuromuscular blockade and their activity outside the neuromuscular ...

Preclinical pharmacology of docetaxel
European Journal of Cancer, 1995Docetaxel is a taxoid cytotoxic agent which promotes tubulin assembly into microtubles and inhibits their depolymerisation. In vitro, docetaxel reduces murine and human tumour cell survival by 50% at concentrations of 4-35 ng/ml, with a greater cytotoxic effect on proliferating than on non-proliferating cells. In vivo, docetaxel is schedule-independent.

Cardiovascular preclinical imaging
The Quarterly Journal of Nuclear Medicine and Molecular Imaging, 2017Non-invasive imaging in the form of single-photon emission-computed tomography (SPECT), positron-emission tomography (PET), computed tomography (CT), echocardiography or magnetic resonance imaging (MRI) is a very useful tool for cardiovascular research as it allows assessment of biological processes in vivo.

Preclinical pharmacology of alendronate
Osteoporosis International, 1993This brief review summarizes some of the preclinical findings of studies aimed at assessing the efficacy and safety of the aminobisphosphonate alendronate (ALN) in preventing or restoring the bone loss caused by calcium or estrogen deficiency. Mode of action studies show that ALN localizes at sites of bone resorption and inhibits osteoclastic activity.

Preclinical evaluation of lovastatin
The American Journal of Cardiology, 1988Administration of lovastatin to animals at high dosage levels produces a broad spectrum of toxicity. This toxicity is expected based on the critical nature of the target enzyme (HMG CoA reductase) and the magnitude of the dosage levels used. The information reviewed in this paper demonstrates that these adverse findings in animals do not predict ...

Preclinical Development of AS04
2009Recent knowledge on vaccine-induced immunity led to the development of vaccine Adjuvant Systems specially designed and adapted to vaccine needs. AS04 is such a tailored Adjuvant System developed by GlaxoSmithKline Biologicals. This chapter focuses on the methods that were used during the preclinical evaluation of AS04.

Antiandrogens are effective drugs in the treatment of advanced prostatic cancer. The main problems associated with their use are their short half-lives, leading to fluctuating serum levels, and their non-selectivity, which leads to increases in serum LH and testosterone through centrally-mediated actions.
