Mosaic chromosomal alterations and relation to haematological cancer in two large Danish cohorts

open access: yesBritish Journal of Haematology, EarlyView.
Summary Mosaic chromosomal alterations (mCAs) are large somatic gains, losses and copy‐neutral loss of heterozygosity identified in the peripheral blood, which represent a relatively unexplored spectrum of clonal haematopoiesis. We aimed to describe mCAs in two large Danish cohorts with extensive long‐term registry‐based follow‐up and examine ...

The lung is more than an organ of gas exchange: immunocompetency, platelets and implications for drug discovery

open access: yesBritish Journal of Pharmacology, EarlyView.
Abstract The lungs have for a long time been recognised as having pharmacological significance as a specific organ to modulate the pharmacokinetics of endogenous substances needed to maintain homeostasis. The lungs are therefore the site of action of certain drugs that act on these homeostatic processes, resulting in systemic effects on the ...

Genomic Evolution and Immune Contexture With Therapeutic Relevance in Pancreatic Neuroendocrine Neoplasms

open access: yesCancer Science, EarlyView.
Lineage‐dependent immunogenomic landscapes and biologically informed therapy in pancreatic neuroendocrine neoplasms. Pancreatic neuroendocrine neoplasms display lineage‐dependent immunogenomic landscapes, in which genomic alterations, epigenetic states, antigen‐presentation status, immune‐cell infiltration, and suppressive microenvironments co‐evolve ...

Monocytes Derived From Human Pluripotent Stem Cells Engineered for Detection of Pyrogens

open access: yesCell Proliferation, EarlyView.
In summary, we have developed a monocyte‐based reporter system using hESC‐Mono for interrogating the NF‐κB signalling axis, a central pathway governing pyrogenic responses. Our platform enables rapid and dose‐dependent detection of both endotoxins (LPS) and non‐endotoxins (LTA and Zymosan).

Integrated Engineering of CAR‐T Cells for Solid Tumours

open access: yesCell Proliferation, EarlyView.
Solid tumours pose multifactorial barriers including antigen heterogeneity, immunosuppressive microenvironment, and poor T‐cell trafficking, limiting CAR‐T efficacy compared to hematologic malignancies. Integrated engineering strategies are essential, combining logic‐gated receptors for precision, metabolic‐epigenetic reprogramming for resilience, and ...

Primary Nodal Poroid Hidradenocarcinoma With YAP1::NUTM1 Fusion: Report of a Rare Case

open access: yesJournal of Cutaneous Pathology, EarlyView.
ABSTRACT Poroid hidradenocarcinoma is a rare malignant sweat gland neoplasm. Although primary nodal hidradenoma has been reported, primary nodal poroid hidradenocarcinoma has not been described. We report a case of a 56‐year‐old man presenting with a nontender 5.5 cm mass within the axillary lymph node, and no identifiable primary cutaneous lesion on ...

The Myeloma Risk Variant of TNFRSF13B Shows Reduced NF‐κB Signalling Activity in HEK293 Cells

open access: yesEuropean Journal of Haematology, EarlyView.
ABSTRACT A single nucleotide variant rs34562254 in TNFRSF13B, which encodes TACI (transmembrane activator and CAML interactor), is associated with an increased risk of myeloma and MGUS. This variant encodes a proline to leucine substitution at residue 251 in the long isoform (P251L) and residue 205 in the short isoform (P205L) of TACI.

Adipocyte‐specific NIK depletion enhances energy metabolism and glucose tolerance in mice

open access: yesThe FEBS Journal, EarlyView.
Obesity is associated with reduced energy expenditure and increased hepatic lipid accumulation. This study reveals that loss of NIK in adipocytes stimulates FGF21‐driven expression of thermogenic genes, including UCP1, and mitochondrial uncoupled respiration.
