Protein tyrosine phosphatase PTPRO represses lung adenocarcinoma progression by inducing mitochondria-dependent apoptosis and restraining tumor metastasis

open access: yesCell Death and Disease
Emerging evidence indicates that protein activities regulated by receptor protein tyrosine phosphatases (RPTPs) are crucial for a variety of cellular processes, such as proliferation, apoptosis, and immunological response.

Discovery of potential protein tyrosine phosphatase non-receptor type 2 inhibitors from phytochemical database through structure- based high-throughput virtual screening and machine learning Discovery of protein tyrosine phosphatase non-receptor type 2 inhibitors

open access: yes
Abstract Protein tyrosine phosphatase non-receptor type 2 (PTPN2) has recently become a viable target for cancer immunotherapy. Our study aimed to predict compounds with potential to inhibit PTPN2 for anticancer immunotherapy through a combination of structure-based and ligand-based virtual screening.

Hypothalamic Control of Liver Health and Disease: From Circuits to Pathophysiology and Therapies

open access: yesAdvanced Science, EarlyView.
This review delineates the hypothalamic circuits that control liver homeostasis via autonomic and neuroendocrine pathways. Dysregulation of this hypothalamus–liver axis drives disease progression across a spectrum including steatotic liver disease, liver inflammation and injury, fibrosis, cirrhosis, and hepatocellular carcinoma.

Genetic Ablation and Multi‐Omics Profiling Reveal CEP55 as a Key Driver of Tumorigenesis in Diverse Cancer Models

open access: yesAdvanced Science, EarlyView.
Genetic ablation of Cep55 in Pten‐deficient mouse models delays tumorigenesis. Integrated multi‐omics analyses (proteomics, phosphoproteomics, and spatial transcriptomics) reveal that CEP55 regulates oncogenic signaling (RAS/ERK, PI3K/AKT), integrin/FAK‐mediated adhesion, extracellular matrix (ECM) remodeling, and endocytosis.

Gαi1/3 Are Essential for BMP2‐Induced Osteogenesis by Bridging BMP Receptors to the Gab1 Signaling Complex

open access: yesAdvanced Science, EarlyView.
The Gαi1/3 protein is a crucial connecting protein closely related to the signal transduction process of bone formation‐related cytokines such as BMP2. Gαi1/3 mediates downstream signaling pathways such as Smads and MAPK by binding to adaptor proteins like Gab1, thereby promoting the osteogenic effect of BMSCs.

B Cells are Activated via a Nanoporous Interface that Stabilizes Microvilli and Engages Mechanosensitive Ion Channels

open access: yesAdvanced Science, EarlyView.
B cells can be activated independently of antigen recognition via mechanical stimulation through nanoporous substrates. Exposure to such substrates induced B cell microvilli extension into the pores, intracellular Ca2+ signaling, phosphorylation of signaling proteins, and CD69 expression.

Loss of SHP1 in Spinal Astrocytes Triggers T‐Lymphocyte Infiltration and Nociceptive Hypersensitivity

open access: yesAdvanced Science, EarlyView.
Astrocyte‐specific SHP1 deletion disrupts astrocyte–vascular interactions and compromises BBB integrity while enhancing STAT1‐dependent CXCL10 expression. These changes synergistically promote peripheral CD4+ and CD8+ T‐cell infiltration into the SDH. The infiltrating T cells secrete IFN‐γ, which in turn activates microglia.

PTPN2 inhibition unleashes response to STING agonism in head and neck squamous cell cancer

open access: yesNature Communications
cGAS-STING signaling can promote antitumor immunity, and tumor cell STING is suppressed in a variety of cancer subtypes that resist immune checkpoint blockade.

METTL7A Downregulation Drives SLC1A5‐Mediated Glutamine Competition to Promote Tumor Proliferation and Suppress CD8+ T Cell Immunity in Gastric Cancer

open access: yesAdvanced Science, EarlyView.
GC cells enhance glutamine accumulation by upregulating SLC1A5 expression. This upregulation not only boosts GC cell proliferation, but also outcompetes CD8+ T cells for glutamine and suppresses their antitumor immunity. Mechanistically, METTL7A deficiency in GC induces SLC1A5 overexpression via an m6A‐dependent pathway and N‐glycosylation ...
