Crystal Structure of the Cul2-Rbx1-EloBC-VHL Ubiquitin Ligase Complex [PDF]
Cullin RING E3 ubiquitin ligases (CRLs) function in the ubiquitin proteasome system to catalyze the transfer of ubiquitin from E2 conjugating enzymes to specific substrate proteins.

ESCRT‐Mimetic Nanodegrader Targets STING for Anti‐Inflammatory Therapy
A nanoplatform‐enabled targeted protein degradation strategy is presented to regulate aberrant STING signaling. STING‐ATTEC induces selective autophagic degradation of STING via formation of a STING–ATTEC–LC3 ternary complex, while the cationic FA‐LNP+ system enhances LC3 generation and targeted delivery. Together, this synergistic approach efficiently

Antrodia cinnamomea reduces obesity and modulates the gut microbiota in high-fat diet-fed mice. [PDF]
BackgroundObesity is associated with gut microbiota dysbiosis, disrupted intestinal barrier and chronic inflammation. Given the high and increasing prevalence of obesity worldwide, anti-obesity treatments that are safe, effective and widely available ...

Bottom-up instructive quality control in the biofabrication of smart protein materials [PDF]
The impact of cell factory quality control on material properties is a neglected but critical issue in the fabrication of protein biomaterials, which are unique in merging structure and function. The molecular chaperoning of protein conformational status

This study explores the opposing effects of the mGluR2 and mGluR3 receptors on amyloid precursor protein processing. mGluR2 promotes amyloidogenic cleavage, while mGluR3 favors non‐amyloidogenic pathways. Using a brain‐penetrant nanobody as a mGluR2 positive allosteric modulator, the study uncovers how its chronic activation aggravates amyloid‐β burden

ARHGEF3 is broadly downregulated across human cancers and correlates with patient prognosis. Tumor‐intrinsic ARHGEF3 activates the RHOA–ROCK–PTEN cascade to inhibit AKT signaling, thereby promoting chemokine‐driven T‐cell infiltration and relieving lipid‐mediated myeloid immunosuppression.

Pathophysiology of acute experimental pancreatitis: Lessons from genetically engineered animal models and new molecular approaches [PDF]
The incidence of acute pancreatitis is growing and worldwide population-based studies report a doubling or tripling since the 1970s. 25% of acute pancreatitis are severe and associated with histological changes of necrotizing pancreatitis. There is still

Unraveling the Role of Linker Design in Proteolysis Targeting Chimeras.
A current bottleneck in the development of proteolysis targeting chimeras (PROTACs) is the empirical nature of linker length structure-activity relationships (SARs). A multidisciplinary approach to alleviate the bottleneck is detailed here. First, we examine four published synthetic approaches that have been developed to increase synthetic throughput ...

Covalent Reprogramming of Kinase Binders to Modulate Protein Abundance
Electrophilic remodeling of a broad‐spectrum kinase binder reveals how subtle chemical changes reprogram protein fate. An acrylamide analog of a multi‐kinase binder selectively stabilizes Aurora kinase A (AURKA) by suppressing its ubiquitination, while a short‐linker variant converts this stabilizer into a degrader.
