Transfection of Cardiac Muscle: Effects of Overexpression of c-myc and c-fos Proto-Oncogene Proteins in Primary Cultures of Neonatal Rat Cardiac Myocytes

open access: closedClinical Science, 1997
1. Studies of cells transfected with specific expression vectors allow functions of specific gene products to be investigated. We first established optimum conditions for transfection of neonatal cardiac myocytes in primary culture. Increased proto-oncogene expression has been implicated in the development of cardiac hypertrophy; we therefore wished to

Regulation of c-fos proto-oncogene expression by growth hormone: protein synthesis is not required for down-regulation

open access: closedEndocrinology, 1993
Growth hormone (GH) has been previously shown in Ob1771 adipose cells to transiently stimulate the expression of the c-fos gene by a protein kinase C (PKC)-dependent pathway. This regulation takes place at a transcriptional level. In the presence of cycloheximide (CHX), stimulation by PKC activators or by serum leads to a "superinduction" of the c-fos ...

Acetaldehyde induces c-fos and c-jun proto-oncogenes in fat-storing cell cultures through protein kinase C activation.

open access: closedAlcohol and alcoholism (Oxford, Oxfordshire), 1994
Hepatic fibrosis is an important morphological feature of alcohol-induced liver injury. We previously reported that acetaldehyde stimulates collagen I and fibronectin gene transcription in rat fat-storing cell (FSC) culture. We here evaluated whether acetaldehyde increases Col I and FN gene transcription through the induction of c-fos and c-jun proto ...

Protein Kinase C Regulates Activation of Mitogen-Activated Protein Kinase and Induction of Proto-oncogene c-fos by Endothelin-1

open access: closedJournal of Cardiovascular Pharmacology, 1992
Endothelins (ETs) are potent regulatory peptides that cause numerous phenotypic changes in glomerular mesangial cells including differential regulation of gene expression and mitogenesis. Although the second messengers produced by activated ET receptors are well characterized, little is known about pathways of nuclear signaling.

Inhibition of proto-oncogene c-fos transcription by inhibitors of protein kinase C and ion transport

open access: closedEuropean Journal of Biochemistry, 1987
Both 4 beta-phorbol 12,13-didecanoate (PDD) and calcium ionophore A23187 induced c-fos mRNA accumulation with similar kinetics in human monocyte-like cells (U937). Their effects were additive. Cells pretreated with PDD were not induced to accumulate c-fos mRNA by PDD but remained responsive to the induction by A23187.

Correlation between lack of bone Gla protein mRNA expression in rat transplantable osteosarcomas and expression of both c‐fos and c‐jun proto‐oncogenes

open access: closedMolecular Carcinogenesis, 1993
AbstractAlkaline phosphatase (AP) activity and expression of bone Gla protein (BGP), c‐fos, and c‐jun were compared in two transplantable osteosarcomas with high potentials for metastasis to the lung. The original spontaneous osteosarcoma (SOS) gradually became histologically undifferentiated, losing its osteogenic activity during serial transfer ...

Localization of C-FOS and C-JUN proto-oncogene proteins in rat pleural mesothelial cells by immunofluorescence and confocal scanning laser microscopy

open access: closedProceedings, annual meeting, Electron Microscopy Society of America, 1995
The proto-oncogenes c-fos and c-jun are members of a multigene family known as immediate early genes (IEG) that are transiently expressed in response to a wide variety of environmental stimuli. Homodimeric complexes of jun gene-family products and heterodimeric complexes of the jun and fos gene-family products form the transcription factor AP-1, which ...

The hepatitis B virus X protein transactivation of c-fos and c-myc proto-oncogenes is mediated by multiple transcription factors

open access: closed, 1992
We have constructed two expression vectors in order to study the action of the HBV 17 Kd X protein on the c-fos and c-myc promoters. The results show that the promoters contain multiple elements that respond to X protein, suggesting involvement of multiple transcription factors. The exact mechanism of the interaction remains elusive, but our data allow

Inhibition of protein kinase C prevents asbestos-induced c-fos and c-jun proto-oncogene expression in mesothelial cells.

open access: closedCancer research, 1997
Asbestos and the phorbol ester tumor promoter, 12-O-tetradecanoylphorbol-13-acetate (TPA), increase c-fos and c-jun mRNA levels and AP-1 DNA binding activity in rat pleural mesothelial (RPM) cells, a target cell of asbestos-induced mesotheliomas (N. H. Heintz et al., Proc. Natl. Acad. Sci. USA, 90: 3299-3303, 1993).
