Health‐Related Quality of Life and Symptom Severity Among Patients With PIK3CA‐Related Overgrowth Spectrum: A Mixed‐Methods Study to Understand Real‐World Experience With Alpelisib Treatment

open access: yesPediatric Blood &Cancer, EarlyView.
ABSTRACT Background PIK3CA‐related overgrowth spectrum (PROS) includes several rare overgrowth disorders resulting from somatic gain‐of‐function mutations in PIK3CA. Despite treatment advances, including the recent approval of alpelisib for PROS in the United States, literature detailing the patient experience with PROS is limited.

pH‐mediated activation of the lysosomal arginine sensor SLC38A9

open access: yesFEBS Letters, EarlyView.
Cells monitor nutrient levels via the lysosomal transporter SLC38A9 to activate the mechanistic target of rapamycin complex 1 (mTORC1). This study reveals that SLC38A9 function is regulated by pH. We identified histidine 544 as a critical pH sensor that undergoes conformational changes to control amino acid efflux from lysosomes; therefore, it ...

Interpreter proxy versus healthcare interpreter for administration of patient surveys following arthroplasty: a pilot study.

open access: yesBMC Medical Research Methodology, 2019
Abstract Background: Clinical quality registries and other systems that conduct routine post-discharge surveillance of patient outcomes following surgery may have difficulty surveying patients who have limited proficiency in the language of the healthcare provider.

Cell geometry and membrane protein crowding constrain Escherichia coli growth rate, overflow metabolism, respiration, and maintenance energy

open access: yesFEBS Letters, EarlyView.
The physical dimensions and shape of bacterial cells define the surface area available to acquire nutrients and the volume available for synthesizing proteins and DNA. Here, we use computational systems biology to decode the importance of cell geometry as a major determinant of prokaryotic phenotype, including growth rate and metabolic efficiency. This

Acknowledging the mess

open access: yesArheologia Moldovei
It is nowadays generally well acknowledged that archaeological excavation and documentation methods have a major impact on interpretations of the record. In the last decades, the effort to mitigate potential biases became increasingly visible, generally

Longitudinal genome‐wide aneuploidy measurements in circulating cell‐free DNA to predict lack of benefit from pembrolizumab in patients with metastatic urothelial cancer

open access: yesMolecular Oncology, EarlyView.
Many patients with urothelial cancer do not benefit from treatment with pembrolizumab, while at risk of severe side effects. Changes in the levels of circulating tumor DNA early during treatment, measured by a simple and affordable assay that can be easily implemented in the clinic, can be used as a prognostic tool to identify these patients.

An Integrated Zooarchaeological and Micromorphological Perspective on Midden Taphonomy at Late Neolithic Çatalhöyük

open access: yesOpen Archaeology, 2022
The disposal of cultural material at Çatalhöyük, Turkey (7100–5950 cal BCE) has created substantial midden deposits between buildings and within abandoned houses.

Guidelines for the use and interpretation of palaeofire reconstructions based on various archives and proxies [PDF]

open access: yesQuaternary Science Reviews, 2018
Abstract We present a comparative analysis of fire reconstructions from tree rings and from wood charcoal preserved in forest soils, peat and lake sediments. Our objective is to highlight the benefits and limits of different archives and proxies to reconstruct fire histories.
