Alpha-synuclein in Parkinson’s disease and other synucleinopathies: from overt neurodegeneration back to early synaptic dysfunction

open access: yesCell Death and Disease, 2023
Although the discovery of the critical role of α-synuclein (α-syn) in the pathogenesis of Parkinson’s disease (PD) is now twenty-five years old, it still represents a milestone in PD research.

Alpha-Synuclein Strain Variability in Body-First and Brain-First Synucleinopathies

open access: yesFrontiers in Aging Neuroscience, 2022
Pathogenic alpha-synuclein (asyn) aggregates are a defining feature of neurodegenerative synucleinopathies, which include Parkinson's disease, Lewy body dementia, pure autonomic failure and multiple system atrophy.

Autonomic Dysfunction in α-Synucleinopathies

open access: yesFrontiers in Neurology, 2019
The α-synucleinopathies are a group of neurodegenerative diseases characterized by abnormal accumulation of insoluble α-synuclein in neurons and glial cells, comprising Parkinson's disease (PD), dementia with Lewy bodies (DLB) and multiple system atrophy

Frequent Nocturnal Vocalization in Pure Autonomic Failure [PDF]

open access: yesJournal of International Medical Research, 2008
Nocturnal vocalization is frequent in Parkinson's disease patients with rapid eye movement (REM) sleep behaviour disorder (RBD). We investigated the frequency of nocturnal vocalization and other sleep problems in patients with pure autonomic failure (PAF) and compared the results with idiopathic Parkinson's disease (IPD) and dementia with Lewy bodies (

Stress induced hypotension in pure autonomic failure [PDF]

open access: yesJournal of Neurology, Neurosurgery & Psychiatry, 2006
A 47 year old woman with pure autonomic failure complained of dizziness during emotional stress. Emotional stimuli have not previously been reported to cause hypotension in patients with autonomic failure. In the patient, ambulatory blood pressure recording revealed severe hypotension (50/30 mm Hg) after a stressful event.

Cutaneous Phosphorylated Alpha‐Synuclein in Lewy Body Dementia

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective To determine the test performance of cutaneous phosphorylated alpha‐synuclein (P‐SYN) in dementia with Lewy bodies (DLB), individuals with reduced Montreal Cognitive Assessment (MoCA) and healthy controls. Methods This is the first subgroup analysis of the Synuclein‐One study, a prospective, blinded study evaluating P‐SYN detection ...

Characterization of the symptoms of neurogenic orthostatic hypotension and their impact from a survey of patients and caregivers

open access: yesBMC Neurology, 2018
Background Neurogenic orthostatic hypotension (nOH) results from impaired vasoconstriction due to dysfunction of the autonomic nervous system and is commonly associated with Parkinson disease (PD), multiple system atrophy (MSA), and pure autonomic ...

Synthetic Nanobiology Actuated Lipometabolic Cell Factory for Autologous Tumor Immunotherapy

open access: yesAdvanced Functional Materials, EarlyView.
FA plays a crucial role in the interaction between tumor cells and the tumor microenvironment, especially for the immune response. A biocatalytic immunoenhancement strategy is developed to boost antitumor immunity by FA metabolic orientation to ceramide. Through the design of this delicate catalytic immunoenhancement strategy, the synthetic nanobiology

Pure autonomic failure followed by amyotrophy

open access: yesNeurology, 2007
Features of autonomic dysfunction may develop late in amyotrophic lateral sclerosis (ALS), but pure autonomic failure with subsequent amyotrophy has not been recognized. We report the rare combination of pure autonomic failure followed by gradually progressive, isolated anterior horn cell disease.
