Radioligand assay in reproductive biology

Seminars in Nuclear Medicine, 1975
Radioligand assays have been developed for the principal reproductive steroids and peptide hormones. Specific binding reagents have included antibodies, plasma binders, and intracellular receptors. In each assay, problems of specificity, sensitivity, and nonspecific inhibitors were encountered.

Radioligand Binding Assay

2003
Characterization and cloning of the AT(1) and AT(2) receptors would not have been possible without an assay that could detect and measure the density and affinity of these receptors. The most frequently used, if not the only, methodological approach used to investigate these receptors is the radioligand binding assay performed either in a test tube ...

Radioligand Binding Assays and Their Analysis

2012
Radioligand binding is widely used to characterize receptors and determine their anatomical distribution, particularly the superfamily of seven transmembrane-spanning G protein-coupled receptors for both established transmitters such as endothelin-1 and an increasing number of orphan receptors recently paired with their cognate ligands.

A radioligand-binding assay for adenosine in tissue extracts

Analytical Biochemistry, 1978
Abstract A radioligand-binding assay employing adenine analog-binding protein from rabbit crythrocytes and [ 3 H]adenosine of high specific activity measures less than 1 pmol of adenosine in neutral HClO 4 extracts of tissue. Adenine and its nucleotides interfere with adenosine binding, so are removed by preliminary chromatography on PEI and ...

COMPUTER ANALYSIS OF RADIOLIGAND ASSAY AND RADIOIMMUNOASSAY DATA

Acta Endocrinologica, 1970
ABSTRACT A computer program written in extended BASIC (BIICAC) is described for analysis of radioimmunoassay and competitive protein binding assay data. This method uses logit and log transformations to obtain a linear dose responsive curve, followed by unweighted and iterated weighted least squares regression analysis (similar to probit ...

In Vitro Muscarinic Receptor Radioligand‐Binding Assays

Current Protocols in Pharmacology, 2010
AbstractG‐protein‐coupled muscarinic receptors (mAChRs), of which there are five subtypes (M1‐M5), are attractive drug targets for a number of disorders. Described in this unit are radioligand‐binding assays for defining the selectivity and affinity of chemical agents at the five mAChR subtypes. Detailed methodologies and troubleshooting strategies are

Radioligand Receptor Assay for Urinary and Serum hCG

The Journal of Clinical Endocrinology & Metabolism, 1975
A radioligand receptor assay system for urinary hCG and serum hCG was developed, using 1100-2000 times g fractions of homogenates of pig testis. This method is specific for hCG and LH, and the limit of detection for hCG was 20 mIU, which could be significantly discriminated from zero at the 95% confidence level.

Development and application of a radioligand-receptor assay for thyrotropin

General and Comparative Endocrinology, 1978
Abstract A membrane-bound receptor preparation for bovine thyrotropin (TSH) was obtained from fresh porcine thyroid glands after homogenization in 0.3 M sucrose and differential centrifugation at 10,000 and 100,000g. Highly purified bovine TSH (30–40 IU/mg) was used as standard and was labeled with 125 I by the chloramine-T method for use as ...

Radioligand purification prior to routine receptor assays

Journal of Steroid Biochemistry, 1988
The need to repurify the commercially available radioligands [3H]estradiol and [3H]testosterone before use in routine assays was investigated. Storage of these products for 2 months after delivery led to appreciable degradation of [3H]estradiol compared to [3H]testosterone.

Labeling of dopamine D3 and D4 receptor subtypes in human peripheral blood lymphocytes with [3H]7-OH-DPAT: a combined radioligand binding assay and immunochemical study

open access: yesJournal of Neuroimmunology, 1998
Molecular biology studies have demonstrated that human peripheral blood lymphocytes express dopamine D2-like receptors belonging to the D3 and D4 receptor subtypes, whereas the characterization of these receptors using radioligand binding assay ...
