Abstract Engineering of enzymes without crystal structures and homologous structures is a great challenge for structure-based computational enzyme design. In this study, a computational strategy that combines protein structure prediction, sequence optimization, and molecular dynamics simulation was developed to improve the catalytic efficiency of the

Metabolism and Toxicity Studies Supporting the Safety of Rebaudioside D
Rebaudioside D (Reb D) is one of the several glycosides found in the leaves of Stevia rebaudiana (Bertoni) Bertoni (Compositae) which has been identified as a potential sweetener. The metabolism of Reb A and Reb D was evaluated in various in vitro matrices (simulated gastrointestinal fluids, rat liver microsomes, and rat cecal contents) and through ...

Rebaudioside D is a promising sweetener due to its zero calorie and high sweetness. Here, a transglucosylase gene eugt11 from Oryza sativa was for the first time expressed in Pichia pastoris, and transformant XE-3 showed the highest expression levels in pH 5.5 BMMY media containing 0.75% methanol.

Separation of rebaudioside D from steviol glycoside by frontal chromatography
Steviol glycoside (SG) is the non-caloric food additive extracted from stevia rebaudiana leaves. It is a mixture of four major components, which are Rebaudioside A (RA), Rebaudioside C (RC), Rebaudioside D (RD) and Stevioside (STV). The RD is capable of providing 250-300 times sweetness than the sugar and giving the clean aftertaste.

In this study, a monoglucosyl rebaudioside A product was isolated from the mixture of glucosylated rebaudioside A obtained from the most reported and industrial used cyclodextrin glycosyl transferase, Toruzyme 3.0 L (CGTase, Toruzyme 3.0 L). The molecular structure of the monoglucosyl rebaudioside A was characterized using LC-MS/MS and methylation ...

The hydrolysis of the steviol glycosides rebaudioside A, B, D, and M, as well as of steviolbioside (a metabolic intermediate) to steviol was evaluated in vitro using human fecal homogenates from healthy donors under anaerobic conditions. Incubation of each of the rebaudiosides resulted in rapid hydrolysis to steviol. Metabolism was complete within 24h,

Synthesis of rebaudioside D, using glycosyltransferase UGTSL2 and in situ UDP-glucose regeneration
Steviol glycosides from Stevia rebaudiana leaves are used in stevia-based sweeteners for their intense sweetness and low calories. Rebaudioside D is present in leaves in minute quantities (∼0.4-0.5% w/w total dry weight), but it is ∼350 times sweeter than sucrose, and sweeter than the more abundant rebaudioside A and stevioside.

Rebaudioside M (Reb M), a zero-calorie sweetener with high sweetness, faces production challenges due to its low yield and purity. UGT76G1, a uridine diphosphate glucose (UDPG)-dependent glycosyltransferase, forms a β-1,3-glycosidic bond with rebaudioside D (Reb D) to produce Reb M but with an efficiency lower than that for stevioside (ST).

Rebaudioside (Reb) D is a high intensity, natural sweetener that shows great potential for substituting sugar in sweetened beverages. However, Reb D is poorly water soluble, and thus, a solid dispersion technique was recently established to enhance its solubility.
