Unveiling the Complete Biosynthetic Pathway of Azacitidine: A Rare 1,3,5‐Triazine Nucleoside Derived From GTP

open access: yesAngewandte Chemie, EarlyView.
The azacitidine biosynthetic gene cluster (aza BGC) was identified by the resistance‐gene‐guided comparative genomics method in Streptomyces mobaraensis IPIO2. The complete biosynthetic pathway of azacitidine, which follows a “base‐first‐then‐glycosylation” logic, was elucidated through in vivo and in vitro experiments.

Noncanonical Peloruside A Biosynthesis by an Uncultivated Verrucomicrobiota Symbiont

open access: yesAngewandte Chemie, EarlyView.
Noncanonical biochemistry of the peloruside PKS characterized in this work, comprising double bond generation by a noncanonical tridomain (blue), acetyl‐CoA recycling (orange), and proofreading systems (purple and yellow). The Z‐double bond is installed by TEB1 through O‐acetylation and acetate elimination. The acyl ligase PelB (AL, orange) selectively

Beyond Nature's Blueprint: Macrocyclic Peptides Containing Unnatural Amino Acids in Therapeutic Development

open access: yesAngewandte Chemie, EarlyView.
Strategic incorporation of unnatural amino acids transforms macrocyclic peptides into drug‐like molecules capable of engaging challenging targets. These building blocks enhance stability, permeability, and bioavailability, accelerating the development of next‐generation peptide therapeutics.

Structure and Biosynthesis of Mangotoxin Produced by the Plant Pathogen Pseudomonas syringae

open access: yesAngewandte Chemie, EarlyView.
The structure of mangotoxin remained elusive for over two decades. Here, mangotoxin was isolated from the native producer, Pseudomonas syringae pv. syringae, and was determined to be an alkyne‐containing dipeptide. Mangotoxin inhibited the growth of multidrug‐resistant human pathogen Klebsiella pneumoniae.

Supramolecular Cadmium‐Telluride Proteoliposomal Assemblies for Real‐Time Detection of Protein‐Catalyzed Pt(II)‐Complexes Transmembrane Translocation: Cu(I)‐Pumps Are Electrogenic Pt(II)‐Drug Uniporters

open access: yesAngewandte Chemie, EarlyView.
Supramolecular mercaptosuccinic acid‐capped CdTe core‐type quantum dot‐encapsulated proteoliposome assemblies (MSA‐CdTe QD PLs) have been developed to study and unambiguously demonstrate real‐time transporter‐mediated turnover translocation of Pt(II)‐based complexes by transmembrane Cu(I) P‐type ATPase pumps (CopA).
