Human renin gene of renin-secreting tumor

A large amount of renin mRNA was found to be expressed in the juxtaglomerular cell (JGC) tumor, as determined by Northern analysis. We have isolated the long 5'-flanking region of the human renin gene from the tumor, and characterized the promoter region with respect to nucleotide (nt) sequence and mRNA transcription start point.

Renin is the initiative enzyme for the renin-angiotensin system. It is synthesized mainly in the juxta-glomerular apparatus and secreted in to the circulation in response to hypotension and hypernatremia. Renin cleaves angiotensin I (Ang I) from angiotensinogen by recognizing the [HPF]-domain on Ang I.

[From the renin gene to renin inhibitors].

The only known action of renin is the hydrolysis of angiotensinogen into angiotensin I. Renin is synthesized as an inactive precursor, preprorenin. The processing of prorenin into active renin occurs after the clivage of a profragment, just after a dibasic pair of amino-acids.

Although the brain contains cathepsins at high concentrations which exhibit a non-specific renin-like activity at acidic pH, the presence of specific renin in the brain has been demonstrated by characterizing its specific properties. Renin was separated from cathepsin by affinity chromatography on casein-Sepharose.
