The role of miR‐335‐5p in the redifferentiation of BRAF p.V600E thyroid cancers

open access: yesMolecular Oncology, EarlyView.
The BRAF p.V600E mutation promotes thyroid cancer dedifferentiation and radioiodine resistance. Using a network approach, we identified miR‐335‐5p as a key regulator of BRAF‐mutated thyroid tumors. Restoring miR‐335‐5p increased thyroid‐specific gene expression and iodine uptake in cells and organoids.

IMPDH inhibition enhances cytarabine efficacy in SAMHD1‐expressing leukaemia cells via guanine nucleotide depletion

open access: yesMolecular Oncology, EarlyView.
Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.

Discrete-continuous analysis of optimal equipment replacement [PDF]

open access: yes
In Operations Research, the equipment replacement process is usually modeled in discrete time. The optimal replacement strategies are found from discrete (or integer) programming problems, well known for their analytic and computational complexity.

A simple method for estimating inter-generational replacement based on fertility and migration European examples

open access: yes, 2010
Concern over unprecedentedly low levels of childbearing in Europe has become increasingly marked among both scientists and policy-makers. In conjunction with these concerns over fertility, there has also been considerable debate on the role migration can

Finding novel vulnerabilities of hypomorphic BRCA1 alleles

open access: yesMolecular Oncology, EarlyView.
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

Introducing AI in Pension Planning: A Comparative Study of Deep Learning and Mamdani Fuzzy Inference Systems for Estimating Replacement Rates

open access: yesMathematics
Funded pensions have become a key focus in strategies to ensure supplementary income during retirement. This paper explores two distinct approaches for estimating replacement rates: a deep learning model and a Mamdani Fuzzy Inference System (FIS).

Excitatory, inhibitory and structural plasticity produce correlated connectivity in random networks trained to solve paired-stimulus tasks

open access: yesFrontiers in Computational Neuroscience, 2011
The pattern of connections among cortical excitatory cells with overlapping arbors is non-random. In particular, correlations among connections produce clustering – cells in cliques connect to each other with high probability, but with lower probability ...

Circulating microRNA signatures of cachexia and cancer in Canis familiaris as a comparative oncology model for human disease

open access: yesMolecular Oncology, EarlyView.
Circulating microRNAs as biomarkers of cachexia and sex‐specific cancer in senior dogs. In 25 client‐owned dogs, four circulating miRNAs (miR‐15a, miR‐15b, miR‐16, miR‐140) were downregulated in cachexia, with miR‐16 the strongest individual biomarker (AUC = 0.899).

Open Cow Replacement Decisions: an Application of Asset Replacement Theory [PDF]

open access: yes
Beef producers must decide what to do with a cow that fails to conceive during the breeding season. Keeping the open cow results in a years expenses without any revenue.

From static archetypes toward dynamic, condition-aware archetypes in UBEM: an integrative review with an illustrative case demonstration

open access: yesFrontiers in Energy Research
Urban Building Energy Modeling (UBEM) is often used for large-scale renovation planning. However, UBEM traditionally uses static archetypes, which fail to reflect the current state of buildings, particularly in partially renovated stocks.
