Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Idiosyncratic Volatility and Expected Returns at the Global Level
The author investigated the existence and significance of a global cross-sectional relation between idiosyncratic volatility and expected returns by introducing a global idiosyncratic volatility measure and globally diversified test assets. He found that portfolios with the highest and lowest global idiosyncratic volatility do not earn significantly ...

Screening and epitope characterization of Nidogen‐2‐specific nanobodies
Camel immunization and phage display were employed to generate high‐affinity VHH nanobodies against Nidogen‐2. After library construction, biopanning, ELISA screening, sequencing, and recombinant expression, selected nanobodies were purified and characterized, leading to the preliminary exploration of a nanobody‐based sandwich ELISA for specific ...

RISK FACTORS PREVENTING RETURN TO BASELINE ACTIVITY LEVEL AFTER BANKART REPAIR
Background As sports specialization continues to grow amongst youth, the prevalence of shoulder instability continues to increase. Bankart repair exists as a primary method to combat these issues, yet a third or more of patients experience recurrent ...

Estimation of Return Levels with Long Return Periods for Extreme Sea Levels by the Average Conditional Exceedance Rate Method [PDF]
Estimation of so-called return levels for environmental extremes is of importance for risk assessment. A particular challenge is to find estimates corresponding to long return periods, as uncertainties in the form of confidence intervals became too wide for practical use when applying conventional methodology where large portions of data are not used ...

Mass spectrometry based identification of AMP‐O‐Tris generated by Thermococcus onnurineus Cas10
Isolated Thermococcus onnurineus Cas10 generates the noncanonical ATP‐derived product AMP‐O‐Tris while in Tris‐containing buffer as identified via mass spectrometry, revealing relaxed nucleophile selectivity under isolated conditions. These findings suggest that multiprotein Csm complex assembly restricts Cas10 reactivity toward canonical cyclic ...

Return to Sport After Proximal Hamstring Tendon Repair: A Systematic Review
Background: Previous studies have evaluated functional outcomes and return-to-sport rates after proximal hamstring tendon (HT) repair. Purpose: To systematically review the literature in an effort to evaluate return-to-sport rates after proximal HT ...
