Syntheses of some fluorescent ribonucleotides [PDF]

open access: yesNucleic Acids Research, 1980
Synthesis of guanosine 3'-phosphodiesters of 1, N6-ethenoadenosine-2-sulfonate or 1, N6-etheno-2-azaadenosine can be done readily with ribonuclease N1. There is a 10-fold difference in quantum yield between the phosphodiesters and the starting fluorescent nucleosides.

Synergistic Killing of Polymyxin B in Combination With the Antineoplastic Drug Mitotane Against Polymyxin-Susceptible and -Resistant Acinetobacter baumannii: A Metabolomic Study

open access: yesFrontiers in Pharmacology, 2018
Polymyxins are currently used as the last-resort antibiotics against multidrug-resistant Acinetobacter baumannii. As resistance to polymyxins emerges in A. baumannii with monotherapy, combination therapy is often the only remaining treatment option.

Reduction of Ribonucleotide Reductase Subunit RRM2 Potentially Impairs Gut Function of Woody Breast Broilers

open access: yesAnimal Research and One Health, EarlyView.
Woody breast compromises meat quality leading to reduced consumer appeal. Although its causes are unclear, improvements observed with certain dietary supplements suggest that gut health may influence woody breast development. Ribonucleotide reductase subunit RRM2 is vital for mitochondrial function and gastrointestinal integrity, and alteration in its ...

Hydroxyurea-Mediated Cytotoxicity Without Inhibition of Ribonucleotide Reductase

open access: yesCell Reports, 2016
In many organisms, hydroxyurea (HU) inhibits class I ribonucleotide reductase, leading to lowered cellular pools of deoxyribonucleoside triphosphates. The reduced levels for DNA precursors is believed to cause replication fork stalling. Upon treatment of

Expression of a pathogenic mutation of SOD1 sensitizes aprataxin-deficient cells and mice to oxidative stress and triggers hallmarks of premature ageing [PDF]

open access: yes, 2014
Aprataxin (APTX) deficiency causes progressive cerebellar degeneration, ataxia and oculomotor apraxia in man. Cell free assays and crystal structure studies demonstrate a role for APTX in resolving 5'-adenylated nucleic acid breaks, however, APTX ...

A Michaelis‐Arbuzov‐Type Pathway to a Protected 2ʹ‐Deoxy‐2ʹ‐Selenomethyl‐Adenosine‐3ʹ,5ʹ‐Phosphoroselenolate Guanosine Dinucleotide for Use in Modified m7G Cap Synthesis

open access: yesChemistry – A European Journal, EarlyView.
Here we report a synthesis of the dinucleotide 4 for a projected future total synthesis of the m7G cap 1. Highlights of the route include: 1) a 3ʹ‐H‐phosphonate/5ʹ‐selenocyanate Michaelis‐Arbuzov coupling to forge the 3ʹ,5ʹ‐phosphoroselenolate; 2) an Amosova reductive selenomethylation to install the 2ʹ‐SeMe; 3) a solid CPR II 5ʹ‐O‐phosphitylation; and

Rational Design Strategies for Stimuli‐Responsive DNAzymes Using Modified and Artificial Nucleotides

open access: yesChemistry – A European Journal, EarlyView.
Activity control of RNA‐cleaving DNAzymes in response to external stimuli has attracted growing interest for applications in DNA‐based diagnostics, therapeutics, and dynamic DNA nanotechnology. This review summarizes rational design strategies for stimuli‐responsive DNAzymes using modified or artificial nucleotides, including split and caged DNAzymes ...

Quantification and expert evaluation of evidence for chemopredictive biomarkers to personalize cancer treatment. [PDF]

open access: yes, 2017
Predictive biomarkers have the potential to facilitate cancer precision medicine by guiding the optimal choice of therapies for patients. However, clinicians are faced with an enormous volume of often-contradictory evidence regarding the therapeutic ...

The Anti-Sigma Factor MucA of Pseudomonas aeruginosa: Dramatic Differences of a mucA22 vs. a ΔmucA Mutant in Anaerobic Acidified Nitrite Sensitivity of Planktonic and Biofilm Bacteria in vitro and During Chronic Murine Lung Infection [PDF]

open access: yes, 2019
Mucoid mucA22 Pseudomonas aeruginosa (PA) is an opportunistic lung pathogen of cystic fibrosis (CF) and chronic obstructive pulmonary disease (COPD) patients that is highly sensitive to acidified nitrite (A-NO2-).

Reprogramming SREBP1‐dependent lipogenesis and inflammation in high‐risk breast with licochalcone A: A novel path to cancer prevention

open access: yesInternational Journal of Cancer, EarlyView.
What's New? Endocrine drugs repurposed from treatment protocols are the primary medications available for breast cancer (BC) prevention in at‐risk women. Adverse effects, however, significantly minimize uptake and prevention impact. Here, the authors investigated licochalcone A (LicA), a non‐endocrine anti‐inflammatory agent with reported hematologic ...
