Comparative studies on bacterial 5S ribosomal RNA

Abstract The 5S rRNAs of Escherichia coli , Bacillus stearothermophilus , and B. subtilis were isolated and their molecular conformation examined. All three 5S rRNAs were similar with regard to nucleotide chain length, base composition and general configuration.

The involvement of 5S RNA in the binding of tRNA to ribosomes

Abstract The tRNA fragment TpψpCpGp was found to bind to 5S RNA. This binding is ten times increased when a specific 5S RNA-protein complex is used. The ability of TpψpCpGp to bind to the complex could be abolished by selective chemical modification of two adenines in 5S RNA.

WE have sequenced the DNA region immediately preceding the structural gene for the 5S ribosomal RNA of yeast (Saccharomyces cerevisiae). Since the 5S RNA, a component of the larger ribosomal subunit, has a 5′ triphosphate1,2, this molecule is likely to be the direct product of the gene, unmodified by maturation at the 5′ end.

SOS ribosomes obtained from E. coli were dissociated into 5s rRNA and -44s particles in which at least one of the protein components was missing. Dissociation was produced by dialysis vs. 0.5 M NH4Cl in Mg++-free medium and the resulting 44s particles were isolated by centrifugation through Mg’+-free sucrose solution.

Metabolism of 5S RNA in the absence of ribosome production

Abstract The results presented in this report show that during early development of Xenopus laevis the synthesis of 5S RNA occurs in blastula embryos, whereas the synthesis of 18S and 28S RNA cannot be detected until gastrulation. Thus the initiation of synthesis of the three ribosomal RNAs is not coordinate during early development.
