RNA-dependent RNA polymerases from flaviviruses and Picornaviridae

Current Opinion in Structural Biology, 2009
Flaviviruses and picornaviruses are positive-strand RNA viruses that encode the RNA-dependent RNA polymerase (RdRp) required for replicating the viral genome in infected cells. Because of their specific and essential role in the virus life cycle, RdRps are prime targets for antiviral drugs.

Structural organization of viral RNA-dependent RNA polymerases

Biochemistry (Moscow), 2013
This review describes available data on the structure of viral RNA-dependent RNA polymerases (RdRP) obtained from X-ray analysis and discusses the functional significance of the structural elements of these enzymes. Because most of the studies done to date relate to RdRP structures of picorna-, flavi-, and caliciviruses, here we consider mostly the ...

Biochemical characterization of rhinovirus RNA-dependent RNA polymerase

Antiviral Research, 2002
Human rhinoviruses (HRV) represent the single most important causative agent of the common cold. The HRV genome encodes an RNA-dependent RNA polymerase (RdRp) designated 3D polymerase that is required for replication of the HRV RNA genome. We have expressed and purified recombinant HRV-16 3D polymerase to near homogeneity from Escherichia coli ...

Viral RNA-Dependent RNA Polymerases: A Structural Overview

2018
Most emerging and re-emerging human and animal viral diseases are associated with RNA viruses. All these pathogens, with the exception of retroviruses, encode a specialized enzyme called RNA-dependent RNA polymerase (RdRP), which catalyze phosphodiester-bond formation between ribonucleotides (NTPs) in an RNA template-dependent manner.

Hepatitis C Virus RNA-Dependent RNA Polymerase (NS5B Polymerase)

2003
The hepatitis C virus (HCV) chronically infects approx 4 million patients in the United States alone, and constitutes a major cause of chronic liver disease and hepatocellular carcinoma (1-3). Current antiviral therapies for chronic hepatitis C remain relatively ineffective and have significant side-effects in many patients.

Structure-Based Inhibition of Norovirus RNA-Dependent RNA Polymerases

Journal of Molecular Biology, 2012
Caliciviridae are RNA viruses with a single-stranded, positively oriented polyadenylated genome, responsible for a broad spectrum of diseases such as acute gastroenteritis in humans. Recently, analyses on the structures and functionalities of the RNA-dependent RNA polymerase (RdRp) from several Caliciviruses have been reported. The RdRp is predicted to
