Purification of Large “Troublesome” Polypeptides by RP-HPLC
Cold Spring Harbor Protocols, 2006INTRODUCTIONThis protocol addresses the group of large polypeptides that have proved to be troublesome to handle because of their low solubility or the presence of secondary structural elements that favor supramolecular self-self assembly. Included in this group are polypeptides with amphipathic α-helical or β-sheet structures with extensive runs of ...

Packing Capillary Columns for RP-HPLC
Cold Spring Harbor Protocols, 2006INTRODUCTIONCapillary or microcolumn (<0.5-mm internal diameter [I.D.]) high-performance liquid chromatography (HPLC) is an extremely powerful technique for separating small quantities of proteins and peptides. This protocol describes a procedure for adapting conventional HPLC systems to provide accurate low-flow rates (0.4-4 μl/min) and gradients ...

Determination of ceftazidime in plasma by RP‐HPLC and ultraviolet detection
Biomedical Chromatography, 2021AbstractA simple high‐performance liquid chromatography method for the determination of ceftazidime in plasma has been developed. Using an ultrafiltration technique samples were separated by reverse‐phase high‐performance liquid chromatography on a Symmetry C18 4.6 × 250 mm column (5.0 μm) and ultraviolet absorbance was measured at 260 nm.

Determination of tortoside A in Ilicis pubescentis by RP-HPLC
China Journal of Chinese Materia Medica, 2011To establish a RP-HPLC method for the determination of tortoside A in Ilex pubences.Kromasil-C18 (4.6 mm x 250 mm, 5 microm) column was used in HPLC with mobile phase acetonitrite-0.1% H3PO4 (17:83), the column temperature was 30 degrees C, the flow rate was 1 mL x min(-1), the detection wavelength was set at 210 nm, and inject volume was 10 microL ...

Pharmaceutical analysis plays a vital role in determining the qualitative and quantitative composition of materials, essential for understanding sample characteristics. Analytical chemistry rifts into two branches qualitative, which classifies components, and quantitative, which measures them.

Resolution Optimization in Ion-Interaction RP-HPLC. [PDF]
Abstract Since in ion-interaction chromatography the dependence of the retention on the concentration of the ion-interaction reagent (IIR) follows different behaviours for the different analytes, the adjustment of IIR concentration can usefully be used to optimize resolution.

RP-HPLC determination of water-soluble vitamins in honey
Talanta, 2011The assessment and validation of reliable analytical methods for the determination of vitamins in sugar-based matrices (e.g. honey) are still scarcely explored fields of research. This study proposes and fully validates a simple and fast RP-HPLC method for the simultaneous determination of five water-soluble vitamins (vitamin B(2), riboflavin; vitamin ...

Multivariate quality assessment of aged RP-HPLC columns
Journal of Chemometrics, 1996The inner structure of RP-HPLC columns can change ('age') during routine use. The columns may be monitored to check the magnitude and type of the changes. One approach is to investigate, besides the chromatograms, the retention behaviour of a few selected solutes at different mobile phases (markers) during routine use. A distinction can be made between

RP-HPLC Determination of Lipophilicity in Series of Corticosteroids
2021The adrenal cortex secretes hormones called corticosteroids, which are divided into: mineralocorticoids and glucocorticoids. They are used for the controlled treatment of various dermatological diseases, since they have shown good results. Lipophilicity is a physicochemical feature of a compound that describes its ability to dissolves in fats, oils and

Determination of biogenic amines by RP-HPLC of the dabsyl derivates
Zeitschrift fr Lebensmittel-Untersuchung und -Forschung, 1996An automated derivatization with dabsyl chloride in combination with a high-performance liquid chromatographic analysis is described for the determination of biogenic amines in complex matrices. The sample clean-up procedure consists of an ultrafiltration step, resulting in average recoveries for cheese in the range of 88% to 100%.
