Enzymatic synthesis of bioactive quinolones and (thio)coumarins by fungal type III polyketide synthases

open access: yesThe FEBS Journal, EarlyView.
Quinolones are valuable scaffolds for drug discovery but are rare in nature. Here, we show that two fungal enzymes, AthePKS and FerePKS, can generate 2‐quinolones and two additional heteroaromatic scaffolds. Using AthePKS, we designed an artificial enzymatic cascade towards an antimicrobial quinolone from a simple precursor and implemented it in E ...

Doping In Vivo Alkylation in E. coli by Introducing the Direct Sulfurylation Pathway of S. cerevisiae

open access: yesAngewandte Chemie, Volume 137, Issue 2, January 10, 2025.
S‐Adenosylmethionine‐dependent methyltransferases are emerging enzymes for selective biocatalytic alkylation. Introduction of the direct sulfurylation pathway from S. cerevisiae improved methylation up to 17‐fold, allowing straight‐forward isolation of the methylated products.
