Calpain small subunit homodimerization is robust and calcium‐independent

Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Mitochondrial remodeling shapes neural and glial lineage progression by matching metabolic supply with demand. Cytoplasmic p21 promotes stemness of colon cancer cells via activation of the NFκB pathway

Cytoplasmic p21 promotes colorectal cancer stem cell (CSC) features by destabilizing the NFκB–IκB complex, activating NFκB signaling, and upregulating BCL‐xL and COX2. In contrast to nuclear p21, cytoplasmic p21 enhances spheroid formation and stemness transcription factor CD133.
