Structure and Function of a Multi‐Megadalton Virus‐Like Proteolytic Dodecahedron

open access: yesAngewandte Chemie, EarlyView.
Zuzalysin, a virulence metallopeptidase of the periodontal pathogen Porphyromonas gingivalis, is secreted as a latent, flexible ≈95‐kDa monomer. Calcium triggers cysteine‐switch activation and hierarchical assembly into pentamers, bipentamers, tripentamers, and a ≈5.6‐MDa, ≈355‐Å dodecahedron. Structures at 1.8–3.6 Å reveal a virus‐sized catalytic cage

A Comprehensive In Vitro Approach for Assessing the Physiological Degradation of Medical Implants: The Silkothane Arteriovenous Graft Case Study

open access: yesAdvanced NanoBiomed Research, EarlyView.
Evaluating the degradation of absorbable medical devices in vitro demands physiologically relevant conditions. This study presents a comprehensive framework applied to a three‐layered silk fibroin/polyurethane arteriovenous graft, employing human enzymes at physiological concentrations over 6 months. Multiparametric analyses of both graft structure and

Sequential Design of Activatable Photosensitizers for Ablation of Glioma Cells in Brain Cancer Tissues

open access: yesAngewandte Chemie, EarlyView.
Gal‐SeNBD‐oleamide is an activatable photosensitizer (PS) with efficient uptake and selective, β‐galactosidase‐triggered activation in cancer cells, enabling efficient photodynamic ablation of glioma cells directly in brain tissue slices from mouse preclinical models and human glioblastoma patients.

A Ruthenium(CAAC‐5) Olefin Metathesis Catalyst for Bioconjugation

open access: yesAngewandte Chemie International Edition, EarlyView.
The aqueous instability of ruthenium catalysts restricts olefin metathesis in biological media. A Ru(CAAC‐5) complex bearing an aniline handle is reported that remains active under biologically relevant conditions and enables bioconjugation to proteins, peptides, and an artificial metalloenzyme, providing a robust platform for olefin metathesis in ...
