Syntheses and evaluation of a homologous series of aza-vesamicol as improved radioiodine-labeled probes for sigma-1 receptor imaging

Bioorganic & Medicinal Chemistry, 2019
Sigma-1 receptor imaging probes for determining the expression levels are desirable for diagnoses of various diseases and companion diagnoses of therapeutic agents targeting the sigma-1 receptor. In this study, we aimed to develop probes with higher affinity for the sigma-1 receptor.

Lu 28-179 labels a σ2-site in rat and human brain

Neuropharmacology, 2002
1'-[4-[1-(4-Fluorophenyl)-1H-indol-3-yl]-1-butyl]spiro[isobenzofuran-1(3H),4'-piperidine] (Lu 28-179) or Siramesine is a sigma-ligand with high specificity for sigma(2)-sites, has been tritiated to directly label these sites in brain. [(3)H]Lu 28-179 bound in a saturable manner to homogenates prepared from rat brain or human cortices with high affinity

In vivo labeling of sigma receptors in mouse brain with [3H]4‐phenyl‐1‐(4‐phenylbutyl)piperidine

Synapse, 1995
Abstract4‐Phenyl‐1‐(4‐phenylbutyl)piperidine(4‐PPBP) is a very potent ligand for σ (Sigma) receptors. The present study was undertaken to evaluate [3H]4‐PPBPas a radioligand for in vivo labeling of cerebral σ receptors. After intravenous administration of [3H]4‐PPBP to mice, there is high uptake of radioactivity in the brain.

Synthesis and pharmacological evaluation of 11C-labeled piperazine derivative as a PET probe for sigma-2 receptor imaging

Nuclear Medicine and Biology, 2015
Abstract Introduction Both subtypes of sigma (σ) receptors, σ 1 and σ 2 , are over-expressed in many cancers with σ 2 proposed as a biomarker of tumor proliferation. We are interested in developing a high affinity selective σ 2 radioligand for in vivo monitoring of proliferative status of solid tumors and response to anti-cancer therapies.

Rat liver and kidney contain high densities of σ1 and σ2 receptors: characterization by ligand binding and photoaffinity labeling

European Journal of Pharmacology: Molecular Pharmacology, 1994
Rat liver and kidney were investigated for the presence of sigma (sigma) receptor subtypes by radioligand binding with three highly selective sigma probes and by photoaffinity labeling using [3H]azido-di-o-tolylguanidine ([3H]azido-DTG). [3H](+)-Pentazocine, a highly selective sigma 1 probe, bound to sites in liver membranes with Kd = 7.5 nM and Bmax3 =

Radiosynthesis of sigma receptor ligands for positron emission tomography: 11C- and 18F-labeled guanidines.

Journal of medicinal chemistry, 1991
A series of analogues of the potent and selective sigma receptor ligand 1,3-ditolylguanidine (DTG) were synthesized and demonstrated to have high affinity for the sigma receptor as measured by in vitro [3H]DTG displacement studies using guinea pig brain tissue.
