The (Glg)ABCs of cyanobacteria: modelling of glycogen synthesis and functional divergence of glycogen synthases in Synechocystis sp. PCC 6803

open access: yesFEBS Letters, EarlyView.
We reconstituted Synechocystis glycogen synthesis in vitro from purified enzymes and showed that two GlgA isoenzymes produce glycogen with different architectures: GlgA1 yields denser, highly branched glycogen, whereas GlgA2 synthesizes longer, less‐branched chains.

Unsupervised spatially embedded deep representation of spatial transcriptomics

open access: yesGenome Medicine
Optimal integration of transcriptomics data and associated spatial information is essential towards fully exploiting spatial transcriptomics to dissect tissue heterogeneity and map out inter-cellular communications.

Magnitude and Sign Correlations in Heartbeat Fluctuations

open access: yes, 2001
We propose an approach for analyzing signals with long-range correlations by decomposing the signal increment series into magnitude and sign series and analyzing their scaling properties.

Organ‐specific redox imbalances in spinal muscular atrophy mice are partially rescued by SMN antisense oligonucleotides

open access: yesFEBS Letters, EarlyView.
We identified a systemic, progressive loss of protein S‐glutathionylation—detected by nonreducing western blotting—alongside dysregulation of glutathione‐cycle enzymes in both neuronal and peripheral tissues of Taiwanese SMA mice. These alterations were partially rescued by SMN antisense oligonucleotide therapy, revealing persistent redox imbalance as ...

Production and Comprehension of Prosodic Markers in Sign Language Imperatives

open access: yesFrontiers in Psychology, 2018
In signed and spoken language sentences, imperative mood and the corresponding speech acts such as for instance, command, permission or advice, can be distinguished by morphosyntactic structures, but also solely by prosodic cues, which are the focus of ...

Sign-Full Random Projections

open access: yes, 2018
The method of 1-bit ("sign-sign") random projections has been a popular tool for efficient search and machine learning on large datasets. Given two $D$-dim data vectors $u$, $v\in\mathbb{R}^D$, one can generate $x = \sum_{i=1}^D u_i r_i$, and $y = \sum_ ...

Tau acetylation at K331 has limited impact on tau pathology in vivo

open access: yesFEBS Letters, EarlyView.
We mapped tau post‐translational modifications in humanized MAPT knock‐in mice and in amyloid‐bearing double knock‐in mice. Acetylation within the repeat domain, particularly around K331, showed modest increases under amyloid pathology. To test functional relevance, we generated MAPTK331Q knock‐in mice.

Checkpoint blockade immunotherapy reshapes the high-dimensional phenotypic heterogeneity of murine intratumoural neoantigen-specific CD8+ T cells

open access: yesNature Communications, 2017
Immune checkpoint blockade (ICB) therapies can unleash anti-tumour T-cell responses. Here the authors show, by integrating MHC tetramer multiplexing, mass cytometry and high-dimensional analyses, that neoantigen-specific, tumour-infiltrating T cells are ...

Spatially informed clustering, integration, and deconvolution of spatial transcriptomics with GraphST

open access: yesNature Communications, 2023
Advances in spatial transcriptomics technologies have enabled the gene expression profiling of tissues while retaining spatial context. Here the authors present GraphST, a graph self-supervised contrastive learning method that learns informative and ...

Scalable ASL sign recognition using model-based machine learning and linguistically annotated corpora [PDF]

open access: yes, 2018
We report on the high success rates of our new, scalable, computational approach for sign recognition from monocular video, exploiting linguistically annotated ASL datasets with multiple signers.
