Indispensable role of Galectin-3 in promoting quiescence of hematopoietic stem cells
Long term haematopoitic stem cells (LT-HSCs) are in a quiescent state during homeostasis, which is critical for their maintenance. Here, the authors show that Gal-3 expression in LT-HSCs is induced in response to Tie2 and Mpl and is both necessary and ...

Model simplification of signal transduction pathway networks via a hybrid inference strategy [PDF]
A full-scale mathematical model of cellular networks normally involves a large number of variables and parameters. How to effectively develop manageable and reliable models is crucial for effective computation, analysis and design of such systems.

Cell surface interactome analysis identifies TSPAN4 as a negative regulator of PD‐L1 in melanoma
Using cell surface proximity biotinylation, we identified tetraspanin TSPAN4 within the PD‐L1 interactome of melanoma cells. TSPAN4 negatively regulates PD‐L1 expression and lateral mobility by limiting its interaction with CMTM6 and promoting PD‐L1 degradation.

Background We previously reported that a vascular endothelial stem cell population resides in pre-existing blood vessels in mice and may contribute to vascular endothelial cells in liver injury or hind limb ischemia models in the long-term.

Mepazine Inhibits RANK-Induced Osteoclastogenesis Independent of Its MALT1 Inhibitory Function
Mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1) is an intracellular cysteine protease (paracaspase) that plays an integral role in innate and adaptive immunity.

We show that the majority of the 18 analyzed recurrent cancer‐associated ERBB4 mutations are transforming. The most potent mutations are activating, co‐operate with other ERBB receptors, and are sensitive to pan‐ERBB inhibitors. Activating ERBB4 mutations also promote therapy resistance in EGFR‐mutant lung cancer.

Background CD157 marks a population of tissue-resident vascular endothelial stem cells (VESCs) in mice known for their critical role in homeostatic endothelial cell (EC) turnover and the rapid response to vascular damage in the liver by regeneration ...

Mechanisms of Ca2+/calmodulin-dependent kinase II activation in single dendritic spines
Activation of Ca2+/calmodulin-dependent kinase II (CaMKII) in dendritic spines is a key step of long-term potentiation (LTP) induction, yet the exact biochemical steps of CaMKIIα activation in dendritic spines remained elusive. In this study, the authors

Tumour–host interactions in Drosophila: mechanisms in the tumour micro‐ and macroenvironment
This review examines how tumour–host crosstalk takes place at multiple levels of biological organisation, from local cell competition and immune crosstalk to organism‐wide metabolic and physiological collapse. Here, we integrate findings from Drosophila melanogaster studies that reveal conserved mechanisms through which tumours hijack host systems to ...

An advanced glioma cell invasion assay based on organotypic brain slice cultures
Background The poor prognosis for glioblastoma patients is caused by the diffuse infiltrative growth pattern of the tumor. Therefore, the molecular and cellular processes underlying cell migration continue to be a major focus of glioblastoma research ...
