Molecular characterization of covRS mutations in M1UK Streptococcus pyogenes

open access: yesFEBS Open Bio, EarlyView.
Group A Streptococcus (GAS) acquires covRS mutations driving a hypervirulent bacterial state, frequently associated with invasive disease‐like necrotizing fasciitis. We demonstrate that the newly emerged M1UK GAS lineage can also acquire these mutations.

Evolution‐guided yeast complementation reveals functional differences in human PSPH variants

open access: yesFEBS Open Bio, EarlyView.
Ancient genomes can help guide which human genetic variants are tested experimentally. This study applies that idea to PSPH, a gene involved in serine biosynthesis, and uses high‐throughput yeast complementation to compare variant function. The findings reveal measurable differences among selected alleles and illustrate the value of evolution‐guided ...

Single-molecule analysis reveals the mechanism of chromatin ubiquitylation by variant PRC1 complexes

open access: yes
Chromatin regulation relies on “writer” enzymes that add posttranslational modifications to histone proteins. Variant polycomb repressive complex 1 (PRC1) exists as several subtypes, which are “writers” of ubiquitylation on histone H2A K118 and K119 ...

Interallelic gene conversion of leukemia-associated single nucleotide variants

open access: yesGene
CRISPR-Cas9 is a useful tool for inserting precise genetic alterations through homology-directed repair (HDR), although current methods largely rely on provision of an exogenous repair template. Here, we tested the possibility of interchanging heterozygous single nucleotide variants (SNVs) using mutation-specific guide RNA, and the cell's own wild-type

Cross-kingdom genomic variation in chicken gut microbiomes: insights from China’s diverse local breeds

open access: yesMicrobiome
Background The gut microbiome possesses substantial genetic diversity that supports microbial adaptation, but the genomic variation patterns across its prokaryotic and viral populations remain incompletely characterized.

Adenosine triphosphate as a modulator of protein interactions and stability

open access: yesFEBS Open Bio, EarlyView.
ATP is best known as the cell's energy currency, but it also shapes how proteins fold, interact, aggregate and form biomolecular condensates. This review explains the emerging physical principles behind these effects, including weak binding to charged protein regions, magnesium‐dependent behaviour and concentration‐dependent control of protein ...

Single nucleotide variants of the SLC2A2 gene in Mexican-origin population

open access: yesGaceta Médica de México
Diabetes, which is caused by environmental and genetic factors, has a high global prevalence (10.5%). Some single nucleotide variants (SNVs) modify the activity of glucose transporters such as GLUT2, which plays a significant role in insulin secretion and glucose metabolism. To analyze the SNVs associated with type 2 diabetes (T2D) in the Mexican ...

Protocol for quantifying miRNA trafficking across the endosomal membrane

open access: yesFEBS Open Bio, EarlyView.
An in vitro protocol measures miRNA uptake into endosomes isolated from mammalian cell extracts, which are free of subcellular contaminants. Performed at 37 °C in the presence of ATP, it ensures the import of single‐stranded miRNA into the endosomal lumen.

Conformational changes in the catalytically inactive nucleotide-binding site of CFTR. [PDF]

open access: yes, 2013
A central step in the gating of the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel is the association of its two cytosolic nucleotide-binding domains (NBDs) into a head-to-tail dimer, with two nucleotides bound at the ...

Accurate single nucleotide variant detection in viral populations by combining probabilistic clustering with a statistical test of strand bias

open access: yesBMC Genomics, 2013
Deep sequencing is a powerful tool for assessing viral genetic diversity. Such experiments harness the high coverage afforded by next generation sequencing protocols by treating sequencing reads as a population sample.
