Bioorganic and Medicinal Chemistry Letters, 2007
A series of beta-aminoamides bearing triazolopiperazines has been prepared and evaluated as potent, selective, orally active dipeptidyl peptidase IV (DPP-4) inhibitors. Efforts at optimization of the beta-aminoamide series, which ultimately led to the discovery of JANUVIA (sitagliptin phosphate, compound 1), are described.

In Vitro Study and Pharmacokinetic Evaluation of Sitagliptin Phosphate Enantiomers in Rat Plasma
Bioanalysis, 2023Background: A chiral HPLC technique was developed to determine sitagliptin phosphate enantiomers in rat plasma in compliance with US FDA regulations. Methods & results: The technique used a Phenomenex column with a mobile phase consisting of a 60:35:5 (v/v/v) blend of pH4, 10-mM ammonium acetate buffer, methanol and 0.1% formic acid in Millipore water.

Efficient stereocontrolled synthesis of sitagliptin phosphate
Tetrahedron: Asymmetry, 2014Abstract The synthesis of sitagliptin phosphate 1 , a novel DPP-IV inhibitor for the treatment of type 2 diabetes mellitus has been accomplished starting from the chiral synthon (1,4-bis[( R )-1-phenylethyl]piperazine-2,5-dione) 2 , involving highly stereocontrolled (>98%) alkylation as a key step, in a good overall yield of 50% over six steps.

Sitagliptin Phosphate Monohydrate
2011Biological target: Sitagliptin is a β-amino acid-derived inhibitor of dipeptidyl peptidase 4 (DPP4), a serine protease with membrane cell-signalling and soluble N-terminal proline peptidase activity. As a result, sitagliptin inhibits the in vivo degradation of the incretins, such as glucagon-like peptide-1 (GLP-1), which are endogenous glucoregulatory ...

Synthesis of Sitagliptin Phosphate by a NaBH4/ZnCl2-catalyzed Diastereoselective Reduction
Chemistry Letters, 2015Abstract A practical asymmetric synthesis of sitagliptin phosphate, from 1-{3-(trifluoromethyl)-5,6-dihydro-[1,2,4]triazolo-[4,3-a]pyrazin-7(8H)-yl}-4-(2,4,5-trifluorophenyl)butane-1,3-dione, in overall 65.3% yield has been reported. The target compound was synthesized via eneamination, diastereoselective reduction, amine-deprotection ...
