Targeting an allosteric site in dynamin-related protein 1 to inhibit Fis1-mediated mitochondrial dysfunction

open access: yesNature Communications, 2023
The large cytosolic GTPase, dynamin-related protein 1 (Drp1), mediates both physiological and pathological mitochondrial fission. Cell stress triggers Drp1 binding to mitochondrial Fis1 and subsequently, mitochondrial fragmentation, ROS production ...

Chromatin Organization Governs Transcriptional Response and Plasticity of Cancer Stem Cells

open access: yesAdvanced Science, EarlyView.
Through a combination of transcriptomic and histone mapping studies integrated with chromatin imaging, we define how chromatin organization in nanoscale packing domains fine tunes the plasticity of cancer stem cells (CSCs) driving evasion from and survival after chemotherapy.

Allosteric regulation of a prokaryotic small Ras-like GTPase contributes to cell polarity oscillations in bacterial motility.

open access: yesPLoS Biology, 2019
Mutual gliding motility A (MglA), a small Ras-like GTPase; Mutual gliding motility B (MglB), its GTPase activating protein (GAP); and Required for Motility Response Regulator (RomR), a protein that contains a response regulator receiver domain, are major

Cell size, mechanical tension, and GTPase signaling in the Single Cell [PDF]

open access: yesarXiv, 2019
Cell polarization requires redistribution of specific proteins to the nascent front and back of a eukarytotic cell. Among these proteins are Rac and Rho, members of the small GTPase family that regulate the actin cytoskeleton. Rac promotes actin assembly and protrusion of the front edge, whereas Rho activates myosin-driven contraction at the back ...

A mechano-chemical feedback underlies co-existence of qualitatively distinct cell polarity patterns within diverse cell populations [PDF]

open access: yes, 2016
Cell polarization and directional cell migration can display random, persistent and oscillatory dynamic patterns. However, it is not clear if these polarity patterns can be explained by the same underlying regulatory mechanism. Here, we show that random, persistent and oscillatory migration accompanied by polarization can simultaneously occur in ...

DNA Origami‐Cyanine Nanocomplex for Precision Imaging of KRAS‐Mutant Pancreatic Cancer Cells

open access: yesAdvanced Science, EarlyView.
DNA origami‐cyanine (Do‐Cy) nanocomplexes are synthesized to selectively image KRAS‐mutant pancreatic cancer cells in the highly desmoplastic tumors. This study shows that the Do‐Cy nanocomplexes are preferentially internalized by cancer cells over other stroma cells via KRAS‐dependent macropinocytosis.

Prodrugs of the Archetypal Dynamin Inhibitor Bis‐T‐22

open access: yesChemMedChem, Volume 17, Issue 24, December 16, 2022., 2022
The Bis‐T series of compounds comprise some of the most potent inhibitors of dynamin GTPase activity yet reported, such as Bis‐T‐22 (2). The catechol moieties are believed to limit cell permeability, rendering these compounds largely inactive in cells.

SIRT3‐Mediated Deacetylation of DRP1K711 Prevents Mitochondrial Dysfunction in Parkinson's Disease

open access: yesAdvanced Science, EarlyView.
SIRT3‐mediated deacetylation of DRP1 at K711 resists mitochondrial dysfunction and DA neuronal death in PD, uncovering a treatment target. Abstract Dysregulation of mitochondrial dynamics is a key contributor to the pathogenesis of Parkinson's disease (PD).

Stability analysis of a signaling circuit with dual species of GTPase switches [PDF]

open access: yesarXiv, 2020
GTPases are molecular switches that regulate a wide range of cellular processes, such as organelle biogenesis, position, shape, and signal transduction. These enzymes operate by toggling between an active ("ON") guanosine triphosphate (GTP)-bound state and an inactive ("OFF") guanosine diphosphate (GDP)-bound state; such a toggle is regulated by GEFs ...

Measuring irreversibility from learned representations of biological patterns [PDF]

open access: yesarXiv, 2023
Thermodynamic irreversibility is a crucial property of living matter. Irreversible processes maintain spatiotemporally complex structures and functions characteristic of living systems. In high-dimensional biological dynamics, robust and general quantification of irreversibility remains a challenging task due to experimental noise and nonlinear ...
