Anti‐PD‐1 Nanobody‐Armored MSLN CAR‐T Therapy for Malignant Mesothelioma: Preclinical and Clinical Studies

A novel therapy using engineered immune cells (NAC‐T cells) showed promise for refractory malignant mesothelioma. Based on the encouraging preclinical data, the first‐in‐human trial is initiated, demonstrating tolerable safety and promising anti‐tumor activity (ORR 63.6%, DCR 100%, including one CR).

Lactylation‐Driven YTHDC1 Alleviates MASLD by Suppressing PTPN22‐Mediated Dephosphorylation of NLRP3

In MASLD, YTHDC1 undergoes increased lactylation and ubiquitination, reducing its expression. AARS1 mediates lactylation at lysine 565, while disrupted binding to LDHA further promotes lactylation, suppressing YTHDC1. PDZK1‐ULK1 Axis Triggers Lipophagy to Inhibit Tumor Progression and Sunitinib Resistance in Clear Cell Renal Cell Carcinoma

This study reveals that PDZK1 loss impairs ULK1‐mediated lipophagy, driving lipid droplet accumulation and sunitinib resistance in clear cell renal cell carcinoma. PDZK1 inhibits LEF1 nuclear translocation, enhancing ULK1 transcription. Pharmacological ULK1 activation resensitizes resistant tumors to sunitinib.
