Aptamer‐Targeted PrPC Drives Colorectal Cancer Metastasis via a LYN‐STAT3 Complex and Enables Liquid Biopsy Detection

open access: yesAdvanced Science, EarlyView.
The aptamer WHY‐3E identifies PrPC as a CRC driver. Stabilized by USP18, endocytosed PrPC forms a LYN/STAT3 complex, upregulating MSN transcription to promote metastasis. Crucially, WHY‐3E sensitively detects PrPC‐positive circulating exosomes, establishing a robust theoretical foundation for non‐invasive clinical diagnostics.

Role of Plasmodium falciparum thrombospondin-related anonymous protein in host-cell interactions

open access: yesMalaria Journal, 2008
Background Thrombospondin-related anonymous protein (TRAP) is essential for sporozoite motility and for liver cell invasion. TRAP is a type 1 membrane protein that possesses multiple adhesive domains in its extracellular region.

Spatial cycles mediated by UNC119 solubilisation maintain Src family kinases plasma membrane localisation

open access: yes, 2017
The peripheral membrane proto-oncogene Src family protein tyrosine kinases relay growth factor signals to the cytoplasm of mammalian cells. We unravel the spatial cycles of solubilisation, trapping on perinuclear membrane compartments and vesicular ...

Activation of Src family tyrosine kinases by ferric ions [PDF]

open access: yes, 2014
The Src-family tyrosine kinases (SFKs) are oncogenic enzymes that contribute to the initiation and progression of many types of cancer. In normal cells, SFKs are kept in an inactive state mainly by phosphorylation of a consensus regulatory tyrosine near ...

IKKβ and USP28 Regulate HEY1 Stability to Promote Cancer Stemness and Immune Evasion in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
The uncovered IKKβ‐USP28‐HEY1 axis fuels cancer stemness and immune evasion in hepatocellular carcinoma. USP28 deubiquitinates HEY1 upon IKKβ‐mediated phosphorylation, conferring PD‐1/PD‐L1 blockade resistance. Pharmacological inhibition of USP28 sensitizes resistant tumors to anti‐PD‐1 immunotherapy, revealing a promising therapeutic strategy ...

Heat Shock Protein 90: From Molecular Chaperone Function to Therapeutic Targeting in Malignancies

open access: yesAdvanced Science, EarlyView.
In this review, an integrated conceptual framework linking HSP90's molecular chaperone functions to its pathological roles in cancer is proposed. HSP90 serves as a central node that integrates oncogenic signaling, buffers proteotoxic stress, maintains cancer stem cell plasticity, and shapes tumor‐immune interactions, all of which converge to drive ...

Structural basis for selective inhibition of Src family kinases by PP1

open access: yes, 1999
BackgroundSmall-molecule inhibitors that can target individual kinases are powerful tools for use in signal transduction research. It is difficult to find such compounds because of the enormous number of protein kinases and the highly conserved nature of

Preparation, Component Identification, and Antioxidant Activity of Anthocyanins From Punica granatum “Tunisia”

open access: yesAgriFood: Journal of Agricultural Products for Food, EarlyView.
The preparation for Tunisian pomegranate anthocyanin extract. The molecular mechanisms underlying the antioxidant effects of these compounds were investigated using network pharmacology and molecular docking. Antioxidant activity was evaluated through DPPH, ABTS, and FRAP assays, whereas the effects on oxidative stress and inflammation were examined in

The Indole Phytoalexin Derivatives Induced a Significant Inhibition on Src Kinase Activity of Human Cancer Cells

open access: yesProceedings, 2019
The Src, a protein kinase, is a family of protein tyrosine kinases (SFKs), and this protein catalyses the phosphorylation of tyrosine. The studies have revealed its key roles in regulating signal transduction from cell surface receptors.
