GABAergic Neuron Activation in the RMTg‐VTA Pathway Modulates Dopaminergic Neuron Excitability and Social Stress Susceptibility in Male Mice

In susceptible mice, dopamine neurons in the VTA exhibit increased excitability along with upregulation of GABAA receptors. Administration of the GABAA receptor agonist muscimol reduces this neuronal hyperexcitability and reverses social avoidance behavior.

The mTOR/GLT‐1 pathway abnormality induces α‐synucleinopathy, contributing to seizures in FCD lesions, which were rescued by treatment with the mTOR inhibitor Rapa or the GLT‐1 enhancer Cef. Intracerebroventricular α‐syn administration attenuated PTZ‐induced seizures in FCD models.

Our previous study suggests that sumatriptan‐induced chest discomfort is associated with hypersensitivity of CSLV afferents via 5‐HT1B/1D receptors on nerve endings. The present study demonstrates that inhaled 5‐HT1B/1D receptor antagonism suppresses this hypersensitivity without compromising antimigraine efficacy, supporting local pulmonary antagonism
