Steroids XXIII. Synthesis of Some Ring A-Fused Heterocyclic Steroids

Several ring A-fused heterocyclic steroids, which were prepared as potential anabolic agents, are described. These include steroids with triazole, tetrahydropyrazine, and benzothiazine rings fused onto positions 2,3 of a derivative of methyltestosterone.

Cyanoacetamide reacted with 3β-hydroxy-16-hydroxymethylene-5-androsten-17-one to form 1-cyano-2-hydroxy-2(16′-3′β-hydroxy-17′-oxo-5′-androstenyl) propionamide or 3-cyano-4,5-dehydro-6-hydroxy-5,6(16′,17′-3′β-hydroxy-5′-androstenyl)-2-piperidone depending on whether a catalytic or a molecular amount of piperidine was used as the basic condensing agent.

Some steroid heterocycles via enamines

Reactions of steroid enamines with suitably substituted aromatic aldehydes have been used to synthesize steroid derivatives with a heterocyclic system joined to C-2 and C-3. Enamines from 3-keto-steroids and o-hydroxybenzaldehydes yield steroid adducts which contain the fused benzopyran nucleus.
