Synthesis and reactivity of silylmethylcyclopropanes

open access: yes, 2011
PhDSubstituted tetrahydrofurans (THFs) are common structural motifs found in natural products. The biological activity and structural complexity of these compounds makes their efficient construction with controlled regio- and stereochemistry a ...

Cell‐cycle‐specific lesion evolution rather than inhibition of double‐strand‐break repair underpins cisplatin radiosensitization

open access: yesMolecular Oncology, EarlyView.
We analyze cisplatin–DNA adducts (CDAs) and double‐strand breaks (DSBs) in a cell‐cycle‐dependent manner. We find that CDAs form similarly across all cell cycle phases. DSBs arise only in S‐phase. CDAs might not directly impair DSB repair, but S‐phase DSB lesions evolve in the presence of CDAs and disrupt repair in G2, also causing radiosensitization ...

Model Development and Stability Analysis for a Turbocharger Rotor System under Multi-Field Coupled Forces [PDF]

open access: yes, 2012
Automotive turbochargers have been widely applied in vehicles in order to increase the power output of internal combustion engines by increasing the air to fuel ratio entering the piston cylinders.

ESR1 methylation and ESR1 mutations in circulating tumor cells (CTCs) and paired plasma‐cfDNA of advanced breast cancer patients: A feasibility proof‐of‐concept study

open access: yesMolecular Oncology, EarlyView.
Circulating tumor cells (CTCs) and plasma cell‐free DNA (cfDNA) were analyzed to detect ESR1 mutations and methylation in patients with advanced breast cancer. CTC‐derived DNA showed higher sensitivity for mutation detection and revealed complementary genetic and epigenetic alterations, highlighting the added value of CTC analysis for understanding ...

Interpreting the effects of DNA polymerase variants at the structural level

open access: yesMolecular Oncology, EarlyView.
Using MAVISp and molecular dynamics simulations, we analyzed over 60 000 missense variants in POLE and POLD1 from ClinVar, COSMIC, cBioPortal, and saturation mutagenesis. Identified mechanistic indicators, including stability, binding, and long‐range, enable structural interpretation, providing ACMG‐like evidence for possible reclassification of VUS ...

Calculating required substructure damping to meet prescribed system damping levels [PDF]

open access: yes, 2007
Structural synthesis is a method of calculating the transient dynamic response of an assemblage of substructures without explicitly assembling and solving a combined system model. While significant computational advantages are provided by this method,

Developmental programmes drive cellular plasticity, disease progression and therapy resistance in lung adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
This study shows that lung adenocarcinomas exploit developmental branching morphogenesis to acquire a therapy resistant basal‐like tumour cell state. This process was found to be regulated by combined TP53 loss‐of‐function and type‐I interferon signalling, identifying a novel axis for biomarker and therapeutic target discovery.

USP29‐regulated noncanonical stabilization of the hypoxia‐inducible factor‐α in aggressive prostate cancer

open access: yesMolecular Oncology, EarlyView.
We identify USP29 as the only DUB mirroring CA9 expression, a marker of hypoxia and HIF pathway activation associated with PCA aggressiveness. USP29 stabilizes HIF‐1α and HIF‐2α via a noncanonical mechanism that is independent of PHD/pVHL activity yet relies on proteasomal regulation, establishing USP29 as a previously unrecognized regulator of hypoxic

Mixed finite element substructure-subdomain methods for the dynamic analysis of coupled fluid-solid interaction problems

open access: yes, 1996
Based on a variational method adopting the dynamic pressure in the fluid and the acceleration in the solid as arguments in the functional, several substructure-subdomain methods are developed to describe the dynamical behaviour of fluid-structure systems
