The safety and the efficacy of amodiaquine (AQ) alone, AQ plus sulfadoxine-pyrimethamine (SP) (AQ plus SP), and artesunate (ART) plus SP (ART plus SP), three possible alternatives to chloroquine (CQ), were investigated in 379 Rwandan children 6–59 months old with uncomplicated Plasmodium falciparum malaria who visited one urban/peri-urban health center

Abstract 373 subjects occupationally exposed to high levels of malaria transmission were randomised into three groups. Group 1 received one tablet of sulfadoxine-pyrimethamine (SP) (each containing sulfadoxine 500 mg and pyrimethamine 25 mg) twice a week; group 2 received two tablets of sulfadoxine-pyrimethamine (SP) once a week; and group 3 received ...

This paper reports a two-phase study in Bangui, Central African Republic (CAR): first, we assessed the clinical efficacy to chloroquine (CQ), sulfadoxine-pyrimethamine (SP), and amodiaquine (AQ), then we tested the efficacy of two combinations: CQ + SP and AQ + SP.

The Time Distribution of Sulfadoxine-Pyrimethamine Protection from Malaria
Bulletin of Mathematical Biology, 2012Sulfadoxine-pyrimethamine (SP) has been one of the most widely used antimalarial treatments world-wide, and is also used prophylactically in vulnerable populations. In this paper, we develop a mathematical model which allows us to infer the time distribution of SP protection from drug-trial data.

To assist the Peruvian Ministry of Health in modifying the malaria treatment policy for their north Pacific coastal region, we conducted an in vivo efficacy trial of sulfadoxine-pyrimethamine (SP) and SP plus artesunate (SP-AS) for the treatment for uncomplicated Plasmodium falciparum infections.

Treatment of vivax malaria with sulfadoxine-pyrimethamine and with pyrimethamine alone
Transactions of the Royal Society of Tropical Medicine and Hygiene, 1979The effect of pyrimethamine and the combination of pyrimethamine-sulfadoxine (Fansidar) upon the termination of the acute attack of vivax malaria was studied in Thailand. Pyrimethamine was found to be ineffective, providing clearance of parasitaemia in only two of six patients by the end of seven days following treatment.

Safety and Toxicity of Sulfadoxine/Pyrimethamine
Drug Safety, 2007Plasmodium falciparum infection during pregnancy is strongly associated with maternal anaemia and low birth weight, contributing to substantial morbidity and mortality in sub-Saharan Africa. Intermittent preventive treatment in pregnancy with sulfadoxine/pyrimethamine (IPTp-SP) has been one of the most effective approaches to reduce the burden of ...

Sulfadoxine-pyrimethamine embryopathy in Wistar rats.
Kaibogaku zasshi. Journal of anatomy, 1998Pregnant Wistar rats received a therapeutic dose (0.72 mg/kg body weight) of intramuscular sulfadoxine-pyrimethamine on days 5, 12, and 19 of gestation. The treated animals showed elevated neutrophilic lobe average and increased urinary excretion of formiminoglutamic acid (FIGLU) as compared with controls (p < 0.001), confirming folic acid deficiency ...

[Mefloquine and sulfadoxine/pyrimethamine overdose in malaria tropica].
Wiener klinische Wochenschrift, 1993A 39 year-old man with malaria due to Plasmodium falciparum received 3500 mg mefloquine over 3 days, in addition to 3250 mg chloroquine and 175/3500 mg sulfadoxine/pyrimethamine. He developed severe neuropsychiatric symptoms and had to be hospitalized. Treatment with diazepam, haloperidol and thioridazine achieved relief of the severe symptoms after 4 ...
