Novel chemotype NLRP3 inhibitors that target the CRID3-binding pocket with high potency

open access: yesLife Science Alliance
The NLRP3 inflammasome drives various diseases. Here, we describe novel potent and selective NLRP3-targeted inhibitors to bolster pharmacological studies and development of NLRP3-targeted therapies.

Temporal distribution of acute kidney injury–related hospitalizations after initiation of sodium–glucose cotransporter 2 inhibitors: A nationwide claims database analysis in older adults with an independent complementary single‐center eGFR analysis

open access: yesJournal of Diabetes Investigation, EarlyView.
Among 190 older adults who experienced AKI‐related hospitalization after SGLT2 inhibitor initiation, 31.1% were hospitalized within 30 days. In an independent single‐center cohort, sustained early eGFR decline was associated with older age and use of one or two triple‐whammy‐related drug classes.

SGLT2i use and fracture risk compared with DPP4i in patients with T2DM

open access: yesJournal of Diabetes Investigation, EarlyView.
ABSTRACT Introduction The association between sodium‐glucose cotransporter‐2 inhibitor (SGLT2i) use and fracture risk remains inconclusive. Most previous studies have focused on treatment‐naïve patients, despite many patients in routine clinical practice initiating SGLT2i therapy after prior dipeptidyl peptidase‐4 inhibitor (DPP4i) use.

Signaling and sites of interaction for RX-871024 and sulfonylurea in the stimulation of insulin release

open access: yes, 1998
The objective of this study was to compare effects of RX-871024, a compound with imidazoline structure, and the sulfonylurea glibenclamide, representatives of two groups of ATP-dependent potassium channel (KATP) blockers, on insulin secretion and ...

Discontinuation of Glucagon‐Like Peptide‐1 Receptor Agonists After Bowel Obstruction or Ileus: A Nationwide, Real‐World, Database Study

open access: yesJournal of Evidence-Based Medicine, EarlyView.
ABSTRACT Objective Although concerns have been raised that glucagon‐like peptide‐1 receptor agonists (GLP‐1RAs) may increase the risk of bowel obstruction or ileus, evidence remains inconclusive. In this context of uncertainty, we examined the frequency and determinants of GLP‐1RA discontinuation after hospitalization for bowel obstruction/ileus ...

Clinical and genetic predictors of secondary sulfonylurea failure in Type 2 diabetes patients: the SUCLINGEN study

open access: yes, 2020
Background: Due to several limitations in the study designs of sulfonylurea pharmacogenomics studies, we investigated the clinical and genetic predictors of secondary sulfonylurea failure in Type 2 diabetes patients.

Sulfonylurea Receptor-1: A Novel Biomarker for Cerebral Edema in Severe Traumatic Brain Injury

open access: yes, 2017
OBJECTIVES: Cerebral edema is a key poor prognosticator in traumatic brain injury. There are no biomarkers identifying patients at-risk, or guiding mechanistically-precise therapies.

Burden and Risk Factors of CGM‐Detected Hypoglycemia in Older Adults With Type 2 Diabetes

open access: yesJournal of the American Geriatrics Society, EarlyView.
Among older adults with diabetes using insulin or sulfonylureas, the median time spent in hypoglycemia was 3.4% and ~66% of participants exceeded the target of < 1% time with CGM glucose < 70 mg/dL. ABSTRACT Background Comprehensive data on the burden of biochemical hypoglycemia in older adults with type 2 diabetes are lacking.
