Clade A protein phosphatases of type 2C PP2Cs dephosphorylate B3 RAF kinases at a conserved serine residue critical for ATP binding, thereby maintaining their basal activity. Under stress, RAFs rapidly autophosphorylate this residue for autoinhibition.
