This non‑enzymatic function of DHODH drives sunitinib resistance by competing with TRIM37 to block TRIM28 ubiquitination, thereby stabilizing TRIM28 and activating VEGFA transcription. Disrupting the DHODH–TRIM28 interaction with lisaftoclax restores drug sensitivity.

Effect of isavuconazole on the pharmacokinetics of sunitinib and its mechanism
Background Sunitinib, a newly developed multi-targeted tyrosine kinase inhibitor (TKI), has become a common therapeutic option for managing advanced renal cell carcinoma (RCC).

miR-96-5p targets PTEN to mediate sunitinib resistance in clear cell renal cell carcinoma
A multiple receptor tyrosine kinase inhibitor, sunitinib, is a first-line therapy for clear cell renal cell carcinoma (CCRCC). Unfortunately, it has the major challenges of low initial response rate and resistance after about one year of treatment.

Model illustrating the proposed mechanism by which chemotherapy‐induced downregulation of OGT disrupts SNAP29 O‐GlcNAcylation, promoting STX17‐SNAP29‐VAMP8 SNARE complex assembly and protective autophagy, leading to chemoresistance, which in turn establishes a feedforward loop to perpetuate drug tolerance.

A multimodal fusion framework integrating sequence, atomic, and fragment representations captures drug–target interactions across multiple scales. The model delivers strong predictive performance and enables efficient virtual screening. Applied to hematopoietic progenitor kinase 1 (HPK1), it identifies structurally diverse inhibitors with nanomolar ...

ABSTRACT Background Gastrointestinal stromal tumors (GISTs) commonly metastasize to the liver and peritoneum. Metastases may be detected synchronously at the time of initial diagnosis or develop metachronously after curative‐intent resection. Nonetheless, the prognostic significance of metastasis timing in advanced GIST remains unclear.
